Behavioral and neurochemical changes in animal models of depression and drug addiction

Abstract:

Drogová závislost se klinicky často vyskytuje současně s jiným psychiatrickým onemocněním, včetně deprese. Cílem práce bylo prozkoumat komorbiditu závislosti a deprese z preklinického pohledu na behaviorální i neurochemické úrovni. Výsledky této práce poskytují důkazy o mezolimbických změnách v potkaním modelu deprese, které by mohly objasnit základní behaviorální mechanismy v modelu autoaplikace látek, kde potkani s fenotypem deprese vykazují vyšší příjem drogy ve srovnání s kontrolní skupinou. Pohlavní rozdíly v problematice drogové závislosti prokázané v této práci vyzývají ke specifické strategii léčby závislostí a rozdílnému přístupu v prevenci relapsů u mužů a žen.

Abstract:

Drug abuse is clinically a frequent comorbidity of other psychiatric disorders including depression. The aim of this thesis was to investigate the comorbidity of addiction and depression at behavioural and neurochemical level in rat model. Results of this study provide an evidence of mesolimbic neurotransmitter changes in the rat model of depression which may elucidate behavioural mechanisms underlying intravenous self-administration studies in which rats with depressive-like fenotype were demonstrated to have higher drug intake in comparison to intact controls. The gender differences in the issue of drug addiction , which were demonstrated in this thesis, encourage to a specific addiction treatment strategy and different approach in preventing relapses in men and women.
