Simulation of Optical Aberrations in Confocal Microscopy

Abstract:

V této práci se zabýváme nejproblémovějšími optickými aberacemi a problémy přenosu signálu v konfokální mikroskopii. Práce popisuje simulační balíček CytoPacq s důrazem na přenos signálu a jak jsou simulované popsané aberace. Navrhujeme metodu na měření nerovnoměrného osvitu skutečných fluorescentních vzorků s mikro kuličkami. Naměřené funkce jsou pak použity k simulaci nerovnoměrného osvitu. Dále ukážeme že hypotéza o tom, že rozdíly funkcí osvitu různých barev jsou způsobeny pouze chromatickými aberacemi, neplatí.

Abstract:

We consider the most problematic optical aberrations and signal transmission problems in confocal microscopy. The thesis describes the simulation toolbox CytoPacq with focus on singal transmission and how the described aberration are simulated. We design method to measure uneven illumination of real fluorescent samples containing micro spheres. The measured functions are then used this to simulate the uneven illumination. Moreover, we show that the hypothesis about the difference of the illumination profile between different colors being caused only by the chromatic aberration is not true.
