The role of MMP13 in cardiomyocytes

Abstract:

Matrix metaloproteázy (MMP) je skupina na zinku závislých proteáz, které se aktivně podílejí na remodelaci extracelulární matrix při mnoha fyziologicko-patologických procesech. V této bakalářské práci byly sledovány změny hladin MMP 3, 13 a 14 v sérech a srdečních tkáních za fyziologického stavu a při ischemii myokardu u modelových zvířat. Hladiny MMP byly měřeny metodou ELISA sendvičového typu, a to z důvodu její vysoké selektivity. Hladiny MMP 3 byly pod detekčním limitem kitu ve všech sledovaných vzorcích. Nejvyšší hladiny MMP 13 byly naměřeny v ischemické srdeční tkáni jak oproti hladinám v kontrolní tkáni, tak oproti hladinám MMP 13 v sérech po navozené ischemii. Nejvyšší hladiny MMP 14 byly naměřeny u séra po 5 minutách od rozvoje ischemie a s časem klesaly. Hladiny v séru byly rapidně vyšší než u vzorku ischemické tkáně. Z těchto výsledků je patrné, že MMP 13 a 14 jsou zapojeny do procesů při ischemii myokardu a následné remodelace srdce.

Abstract:

The matrix metalloproteinases (MMP), belonging to the group of zinc-dependent proteases, are active in the remodeling of extracellular matrix during various physiological and pathological processes. The aim of the work was to study the changes of the MMP 3, MMP 13 and MMP 14 levels in sera and in cardiac tissues under physiological condition and in the ischemia of myocardium in experimental animals. The levels of MMP were measured by sandwich ELISA method. The levels of MMP 3 were under the detectable limit in all studied samples. The highest levels of MMP 13 were found in the ischemic cardiac tissue compared to both the level in the control tissue and to levels of sera after ischemia. The highest levels of MMP 14 were observed in the serum after 5 minutes from the development of ischemia with decreasing trend in time. The levels of MMP 14 in serum were significantly higher than in ischemic tissue sample. From these results we can conclude that MMP 13 and 14 are engaged in the processes during ischemia of myocardium and in remodeling of the heart.
