Effect Of Reaction Conditions on Enzyme Activity And Inhibitory Concentration

Abstract:

Tématem této práce byl vliv reakčních podmínek na aktivitu enzymu a inhibiční koncentraci. Teoretická část je věnována cholinesterázám a jejich inhibitorům. Zabývá se popisem struktury cholinesteráz, jejich funkcí a mechanismem katalytického působení a možnostmi ovlivnění jejich aktivity na různých úrovních. Dále je zde popsána řada onemocnění souvisejících s cholinesterázami a v neposlední řadě také jednotlivé analytické metody pro stanovení aktivity a inhibice cholinesteráz. V experimentální části se práce zabývá testováním různých reakčních podmínek za účelem získání optimálních podmínek pro měření aktivity lidských cholinesteráz pomocí sledování změny pH. Práce porovnává metodu měření změny pH s titrační pH-statovou metodou a je také věnována stanovení rozdělovacích koeficientů testovaných inhibitorů v systému n-oktanol:voda.

Abstract:

The aim of this work was evaluating of the influence of reaction conditions on enzyme activity and inhibitory concentration. The theoretical part describes cholinesterases and their inhibitors. It deals with the description of the structure of cholinesterases, their function and mechanism of catalytic action and the possibility of influencing their activity at different levels. Furthermore, a number of cholinesterase-related diseases are described, as well as individual analytical methods for determining cholinesterase activity and inhibition. In the experimental part, the thesis deals with testing of different reaction conditions in order to obtain optimal conditions for measurement of human cholinesterase activity by monitoring pH change. The work also compare the pH change method with the pH-stat titration. At last, the partition coefficient of tested inhibitors in the n-octanol: water system was established.
