Characteristics and origin of mitochondrial DNA lineages of selected populations of the brown frog (Rana temporaria) in Czechia

Abstract:

Studium původu linií mitochondriální DNA populací druhu Rana temporaria v České republice má klíčový význam pro pochopení historie a genetické diverzity těchto populací, neboť se jedná o druh s předpokládaným poklesem populací v blízké budoucnosti, a bude si zřejmě zasluhovat náležitou pozornost a ochranu. Z lokalit Česka jsem pilotně geneticky analyzovala 53 jedinců hnědě zbarvených dospělých skokanů a pulců morfologicky zapadajících do rodu Rana. Ti byli nasbíráni na 12 lokalitách Karpat, Ostravska, Jesenicka a západních Čech. Úspěšně byly osekvenovány PCR fragmenty mitochondriálního DNA genu pro cytochrom b (cyt b) v editované délce 509 bp. Konstrukce haplotypové sítě 48 jedinců druhu Rana temporaria této studie a referenčních haplotypů z databáze GenBank poukázala s výjimkou jednoho jedince na 100% sekvenční podobnost jedinců napříč studovanými lokalitami Karpat, Ostravska, Jesenicka a západních Čech a 100% shodu s publikovaným dominantním haplotypem náležící do východoevropské mitochondriální linie Rana temporaria, kam patří také populace severního Balkánu. Výsledky z geneticky studovaných lokalit Česka zatím naopak nepotvrdily přítomnost západní mitochondriální linie. Pro poslední postglaciální kolonizaci Česka se jako příhodná zdrojová populace jeví právě oblast glaciálních refugií Balkánu či oblasti dnešní Ukrajiny nebo Ruska. Didaktická část práce je věnována návrhu vyučovací hodiny a pracovního listu, který seznamuje žáky a učitele s významem, ohrožením a ochranou žab. Návrh byl úspěšně realizován na dvou gymnáziích a zhodnocen jako zajímavý, použitelný a přínosný.

Abstract:

The study of the origin and variation in the mitochondrial DNA of Rana temporaria populations in the Czech Republic is of key importance for understanding the history and genetic diversity of these populations. This species is with a predicted population decline in the near future, and it will probably deserve our attention and protection. From localities in the Czech Republic, I genetically analyzed 53 individuals of brown frogs adults and tadpoles morphologically belonging to the genus Rana. They were collected at 12 locations in the Carpathians, Ostrava Region, Jeseníky Region and Western Bohemia. PCR fragments of the mitochondrial DNA gene for cytochrome b (cyt b) in edited length of 509 bp were successfully sequenced. The construction of the haplotype network of 48 individuals of Rana temporaria coming from this study and from reference haplotypes received from the GenBank database showed, with the exception of one individual, 100% sequence similarity of the individuals across the Czech studied localities and 100% agreement with the published dominant haplotype belonging to the East European mitochondrial lineage Rana temporaria, which also includes the population of the Northern Balkans. On the other hand, the results from the genetically studied locations in the Czech Republic have not yet confirmed the presence of the western mitochondrial lineage. For the last post-glacial colonization of the Czech Republic, the area of glacial refugia in the Balkans or the regions of today's Ukraine or Russia appears to be a suitable source population. The didactic part of the work is devoted to the design of a lesson and worksheet that introduces students and teachers to the importance, threats and conservation of frogs. The proposal was successfully implemented in two high schools and evaluated as interesting, useful and beneficial.
