The use of near-infrared spectroscopy for the determination of fatty acids composition in rapeseed

Abstract:

Diplomová práce se zabývá využitím metody spektroskopie v blízké infračervené oblasti s Fourierovou transformací (FT-NIR) pro stanovení obsahu hlavních mastných kyselin přítomných v semenech řepky olejné. Vybrané vzorky semen byly nasnímány pomocí FT-NIR spektrometru a poté analyzovány laboratorní referenční metodou plynové chromatografie. Byly doplněny, upraveny, kalibrovány a validovány FT-NIR kalibrační modely pro predikci obsahu hlavních mastných kyselin (kyseliny palmitové, stearové, olejové, linolové, linolenové a erukové) sledovaných v oleji řepky. Bylo zjištěno, že lze vytvořit kvalitní a spolehlivé FT-NIR kalibrační modely pro stanovení obsahu majoritně zastoupených mastných kyselin v semenech řepky olejné. Nejlepší výsledky dosahovaly kalibrační modely pro predikci obsahu kyseliny olejové, linolové a linolenové, které vykazovaly nízký rozptyl hodnot chyb (standardní chyba kalibrace, chyba křížové validace, chyba predikce) a dosti silnou regresní závislost mezi naměřenými NIR spektry a referenčními hodnotami. Spolehlivé kalibrace byly vytvořeny také pro stanovení obsahu kyseliny palmitové a stearové, které vykazovaly nízký rozptyl hodnot chyb, ale zároveň nižší hodnotu korelačního koeficientu kalibrace. Vyšší rozptyl hodnot chyb dosáhl model pro predikci kyseliny erukové, nicméně pro potřeby šlechtění a genofondu lze daný model označit za dostatečně robustní. Predikční kvalitu FT-NIR modelů potvrdily také výsledky t-testu na nezávislém souboru vzorků. Z výsledků t-testu je zřejmé, že nebyl mezi referenčními a NIR hodnotami zjištěn statisticky významný rozdíl na hladině významnosti ? = 0,05. Z dosažených výsledků práce vyplývá, že metoda FT-NIR poskytuje dostatečně spolehlivé výsledky pro stanovení obsahu kyseliny palmitové, stearové, olejové, linolové, linolenové a erukové v řepkovém semeni.

Abstract:

The thesis deals with the use of Fourier transform near-infrared spectroscopy (FT-NIR) for the determination of the main fatty acids composition in rapeseed. The chosen rapeseed samples were scanned by the FT-NIR analyzer and then were analysed by gas chromatography as a reference method. The FT-NIR calibration models for the prediction of the main fatty acids composition observed in rapeseed oil (palmitic acid, stearic acid, oleic acid, linoleic acid, linolenic acid and erucic acid) were completed, calibrated and validated. It was found that it was possible to create quality and reliable FT-NIR calibration models for the prediction of the main fatty acids composition in rapeseed. The calibration models for the prediction of oleic acid, linoleic acid and linolenic acid composition achieved the best results and showed a low variance of error values (standard error of calibration, standard error of cross validation, standard error of prediction) and showed strong enough regression dependence between the measured NIR spectra and the reference values. The reliable calibrations were also created for the determination of palmitic acid and stearic acid composition and showed low variance of error values but also lower value of the correlation coefficient. Higher variance of error values was achieved by the erucic acid prediction model, however for breeding and genofond needs the model can be considered as sufficiently robust. The predictive quality of the FT-NIR models was also confirmed by the results of the t-test on the independent set of samples. From the t-test results it is clear that there was no statistically significant difference between the reference and NIR values at the significance level ? = 0.05. The achieved results show that the FT-NIR method provides sufficiently reliable results for the determination of palmitic acid, stearic acid, oleic acid, linoleic acid, linolenic acid and erucic acid composition in rapeseed.
