Extracellular matrix of human lungs: isolation and morphological, functional, and biochemical characterization

Abstract:

Extracelulární matrix představuje komplexní strukturu tkání, která neslouží pouze jako trojrozměrný podklad pro růst buněk, ale funguje také jako klíčový regulátor jejich chování. Její složení a trojrozměrné uspořádání je specifické pro každou tkáň, ve které se podílí na její funkci. Naše zkoumání bylo zaměřeno na extracelulární matrix distální části plic, která je zodpovědná za přenos kyslíku přes alveolokapilární bariéru, odkud je kyslík dále transportován k cílovým tkáním. Hlavním cílem této diplomové práce bylo izolovat tuto strukturu z distální části plic člověka pomocí metody decelularizace a charakterizovat její morfologii, komponenty a funkční vlastnosti získaného decelularizovaného 3D nosiče. K dosažení tohoto cíle jsme otestovali tři odlišné decelularizační protokoly a zkoumali jejich účinky na získaný 3D nosič. V druhé fázi byla provedena analýza proteomu nativní a decelularizované distální části plic, díky které jsme získali cenné informace o složení této struktury.

Abstract:

The extracellular matrix represents a complex tissue structure that not only serves as a three-dimensional substrate for cell growth, but also acts as a key regulator of their behavior. Its composition and three-dimensional arrangement are specific to each tissue in which they function. Our research was focused on the extracellular matrix of the distal lung, which is responsible for the transfer of oxygen through the alveolocapillary barrier, from where it is further transported to the target tissues. The main goal of this thesis was to isolate this structure from the distal lung of the human using the decelularization method and then to characterize the morphology, components and functional properties of the acquired decellularized 3D scaffolds To achieve this goal, we tested three different decelularization protocols and examined their effects on the 3D scaffolds obtained. In the second phase, there was performed proteomic analysis of the native and decellularized distal lung tissue, which gave us valuable information on the composition of this structure.
