Study of the biogenic amines degradation

Abstract:

Disertační práce se zabývá izolací a identifikací mikroorganizmů degradujících biogenní aminy a následným studiem kinetiky jejich rozkladu. Jednotlivé mikroorganizmy byly získány analýzou komerčně dostupných potravin a potravin vyrobených v potravinářských laboratořích FT UTB. Tyto mikroorganizmy byly následně identifikovány pomocí MALDI-TOF MS. Výsledky identifikace metodou MALDI-TOF byly verifikovány sekvenací části genu pro 16S rRNA. Cílem práce bylo in vitro studium schopnosti získaných a identifikovaných izolátů degradovat biogenní aminy za působení různých kombinací vnějších podmínek a kvantifikace snížení koncentrace biogenních aminů tryptaminu, beta-fenylethylaminu, putrescinu, kadaverinu, histaminu a tyraminu. Základní schopnost degradace byla určena na základě schopnosti růstu v minerálním médiu s biogenními aminy jako jedinými zdroji uhlíku a dusíku. Izolované kmeny se schopností degradace jednoho a/nebo více biogenních aminů byly kultivovány v médiu obohaceném o příslušné biogenní aminy, derivatizovány a separovány za pomoci HPLC. V experimentální části práce bylo analyzováno 895 vzorků potravin, ze kterých bylo izolováno 114 mikroorganizmů degradujících biogenní aminy. Pomocí MALDI-TOF byly izoláty identifikovány. Ze všech testovaných izolátů bylo získáno 22 různých druhů mikroorganizmů. Identifikace 13 vybraných mikroorganizmů byla ověřena pomocí PCR a následnou sekvenací (16S rRNA). U izolátů Acinetobacter pittii, Agrobacterium radiobacter, Bacillus altitudinis, Bacillus pumilus, Bacillus safensis, Bacillus subtilis, Enterobacter cloacae, Klebsiella pneumoniae, Micrococcus luteus, Pseudomonas koreensis, Pseudomonas fulva, Serratia marcescens, Serratia ureylitica byla zjištěna významnější míra degradace biogenních aminů a byla pozorována schopnost degradovat všechny testované biogenní aminy v rozmezí 8-100 %. Nejvyšší míra degradační aktivity byla zaznamenána u kmene Bacillus subtilis IB23, u nějž byl dále sledován vliv vnějších podmínek na degradační aktivitu. Testované parametry kultivace (teplota, pH média, způsob kultivace) výrazně neovlivnily míru degradace a všechny biogenní aminy byly degradovány o více než 50 %. V další fázi práce byl porovnáván vliv vnějších faktorů (teplota, pH a typ média, koncentrace soli) na míru degradace u kmenů Bacillus subtilis IB23 a Bacillus pumilus IB26. Bylo zjištěno, že pH média v testovaném rozsahu výrazně neovlivňuje míru degradace v ani jednom z testovaných typů médií. Rozklad BA probíhal naopak v menší míře se zvyšující se koncentrací soli (0-3 % w/v) a snižující se teplotou. V nutrient broth bylo prokázáno, že míra degradace je závislá pouze na době kultivace při 30 °C. Srovnání degradace mezi kmeny Bacillus subtilis IB23 a Bacillus pumilus IB26 ukázalo, že B. subtilis IB23 efektivněji degraduje BA v minerálním médiu, B. pumilus IB26 naopak v nutrient broth.

Abstract:

In the dissertation thesis we investigate the isolation and identification of microorganisms degrading biogenic amines and the study of the kinetics of their decomposition. Individual microorganisms were obtained by analyzing commercially available foods and foods produced in the food laboratories of FT UTB. These microorganisms were subsequently identified using MALDI-TOF MS. The results of the MALDI-TOF identification were verified by sequencing part of the 16S rRNA gene. The aim of the work was to study the ability of the obtained and identified isolates to degrade biogenic amines under the influence of various combinations of external conditions in vitro and to quantify the decrease in the concentration of biogenic amines tryptamine, beta-phenylethylamine, putrescine, cadaverine, histamine and tyramine. The basic degradability was determined based on the ability to grow in a mineral medium with biogenic amines as the sole sources of carbon and nitrogen. Isolated strains with the ability to degrade one and/or more biogenic amines were cultured in a medium enriched with the appropriate biogenic amines, derivatized with dansyl chloride and separated using HPLC. In the experimental part of the work, 895 food samples were analyzed, from which 114 microorganisms degrading biogenic amines were isolated. Using MALDI-TOF, the isolates were identified. 22 different types of microorganisms were obtained from all tested isolates. The identification of 13 selected microorganisms was verified using PCR and sequencing. In isolates of Acinetobacter pittii, Agrobacterium radiobacter, Bacillus altitudinis, Bacillus pumilus, Bacillus safensis, Bacillus subtilis, Enterobacter cloacae, Klebsiella pneumoniae, Micrococcus luteus, Pseudomonas koreensis, Pseudomonas fulva, Serratia marcescens, Serratia ureylitica, the degradation rate of biogenic amines was observed and the ability degrade all tested biogenic amines in the range of 8-100 % was proved. The highest degree of degradation activity was recorded in the strain Bacillus subtilis IB23, in which the influence of external conditions on degradation activity was monitored. The tested cultivation parameters (temperature, pH medium, cultivation method) did not significantly affect the degradation rate and all biogenic amines were degraded by more than 50 %. In the next phase of the work, the influence of external factors (temperature, pH and type of medium, salt concentration) on the degree of degradation in Bacillus subtilis IB23 and Bacillus pumilus IB26 strains was compared. It was found that the pH of the medium in the tested range does not significantly affect the rate of degradation in any of the tested types of media. On the contrary, BA decomposition was lower with increasing salt concentration (0-3 % w/v) and decreasing temperature. In nutrient broth, it was demonstrated that the rate of degradation is dependent only on the time of cultivation at 30 °C. Comparison of degradation between Bacillus subtilis IB23 and Bacillus pumilus IB26 strains showed that B. subtilis IB23 degrades BA more efficiently in mineral medium, B. pumilus IB26 on the contrary in nutrient broth.
