Determination of internal dynamics of protein MAP2c using cross-correlated NMR relaxation

Abstract:

Mezi všemi proteiny, které jsou nezbytné pro správné fungování buňky, existuje velká část těch, které nemají definovanou 3D strukturu. Jedním z těchto tzv. přirozeně neuspořádaných proteinů je Microtubule Associated Protein 2c který se podílí na regulaci stability a dynamiky mikrotubulů nervových buněk. Funkce přirozeně neuspořádaných proteinů souvisí s přechody mezi velkým počtem nestrukturovaných konformací v nevázané formě a strukturovaných konformací ve vázané formě. V této práci byla použita nukleární magnetická rezonance k popisu vnitřních pohybů, strukturního chování a interakčních vlastností Microtubule Associated protein 2c. Měření chemických posunů, auto- a kros- korelovaných relaxačních rychlostí a zbytkových dipolárních interakcí Microtubule Associated Protein 2c a jeho krátkých fragmentů obsahujících funkčně zajímavé oblasti poskytlo informace o vlivu absence zbývajících domén na vnitřní pohyby a strukturní vlastnosti. Navíc, mapování vazebných míst s rozlišením jednotlivých reziduí odhalilo vazbu plektinu jako potenciální funkci dosud funkčně nepopsaného strukturního motivu.

Abstract:

Among all the proteins that are essential for the right cellular functionality, there is large portion of those, which lack of defined 3D structure. One of these so-called Intrinsically disordered proteins is Microtubule Associated Protein 2c, which is involved in the regulation of the stability and dynamics of microtubules of neural cells. The function of Intrinsically Disordered Proteins arises from the transitions between a large number of free disordered and bound ordered conformations. In this work Nuclear Magnetic Resonance was used to describe local motions, structural behavior and interaction properties of Microtubule Associated Protein 2c. Measurement of chemical shifts, auto- and cross-correlated relaxation rates and residual dipolar couplings of full-length Microtubule Associated Protein 2c and also its shorter fragments containing functionally interesting regions provided information on the effects of absence of other domains to the internal motions and structural properties. Additionally, residue-specific binding site mapping revealed a potential plectin-binding role of so-far functionally uncharacterized structural motif.
