Abstract:

Dietární faktory významným způsobem ovlivňují vznik a vývoj nádorů, a to zejména v případě nádorů gastrointestinálního traktu. V potravinách se může vyskytovat řada nebezpečných látek majících mutagenní vlastnosti či podporujících rozvoj nádorových onemocnění. Jedná se nejen o látky anorganické, jako jsou např. arsen a některé jeho sloučeniny, ale i řadu organických sloučeniny klasifikovaných jako lidské karcinogeny. Mezi ně patří například některé perzistentní halogenované aromatické uhlovodíky (HAU) nebo polycyklické aromatické uhlovodíky (PAU). Mnohé z těchto sloučenin jsou účinnými exogenními ligandy receptoru pro aromatické uhlovodíky (AhR), jehož cílovými geny jsou mimo jiné i enzymy I a II fáze biotransformace xenobiotik. Tyto látky pak aktivují svůj vlastní metabolismus, což může vést nejen k jejich detoxifikaci, ale i bioaktivaci vytvářející reaktivní sloučeniny schopné vazby na buněčnou DNA. Tyto potenciálně nebezpečné látky mohou v gastrointestinálním traktu interagovat nejen s dalšími složkami potravy, ale i s produkty střevního mikrobiomu, což může mít ovlivnit jejich metabolismus a toxicitu, včetně jejich mutagenity. Mezi významné skupiny látek produkovaných střevním mikrobiomem patří zejména mastné kyseliny s krátkým řetězcem, jako jsou acetát, propionát a butyrát. Zejména butyrát je znám tím, že představuje důležitý zdroje energie pro buňky epitelu tlustého střeva a zároveň reguluje procesy proliferace, diferenciace a buněčné smrti v normálních i nádorových buňkách tlustého střeva. O jeho působení na regulaci enzymů metabolizujících a aktivujících karcinogeny ve střevním epitelu se však ví poměrně málo. V rámci této disertace jsou prezentovány práce prokazující, že butyrát může významným způsobem ovlivnit expresi a aktivitu enzymů podílejících se metabolismu karcinogenů jako je benzo[a]pyren (BaP), v řadě buněčných modelů odvozených od epitelu tlustého střeva. Butyrát především posiluje indukci exprese a/nebo aktivity cytochromů P450 rodiny 1 (CYP1), regulovaných AhR a hrajících kritickou roli v metabolismu BaP. Významně tak ovlivňuje produkci metabolitů BaP, včetně tvorby reaktivních metabolitů vytvářejících kovalentní DNA adukty. Tyto účinky butyrátu souvisejí s inhibicí histonových deacetylas (HDAC), ovlivňujících transkripci klíčových cílových genů AhR, jako je právě CYP1A1. Účinky butyrátu nejsou omezené pouze na rodinu enzymů CYP1, ale ovlivňují širší spektrum enzymů podílejících biotransformaci xenobiotik a metabolismu karcinogenů. Jedná se zejména o expresi a aktivitu enzymů jako jsou N acetyltransferasy, NAD(P)H:chinonoxidoreduktasa a UDP-glukuronosyltransferasy, významné enzymy podílející se aktivaci nebo detoxifikaci dietárních karcinogenů. Naše výsledky tak poprvé prokazují, že butyrát, běžný produkt mikrobiální fermentace vlákniny, je schopen významně ovlivnit metabolismus některých toxických dietárních kontaminantů, a ovlivňovat nejen jejich detoxifikaci, ale i tvorbu jejich genotoxických metabolitů v buňkách tlustého střeva.

Abstract:

Both origin and further development of cancer are significantly affected by dietary factors, in particular in the case of colorectal cancer. A number of dangerous substances with mutagenic properties or promoting the development of cancer can be a part of the diet. These include not only inorganic substances, such as arsenic and arsenic-based compounds, but also a number of organic compounds classified as human carcinogens. These include e.g. some persistent halogenated aromatic hydrocarbons (HAHs) or polycyclic aromatic hydrocarbons (PAHs). Many of these compounds are effective exogenous ligands of the aryl hydrocarbon receptor (AhR), which target genes include, among other genes, many phase I and phase II biotransformation enzymes. These compounds can thus activate their own metabolism, which can lead not only to their detoxification but also to their bioactivation and formation of reactive compounds capable of binding DNA. These potentially harmful substances can interact in the gastrointestinal tract not only with other dietary components but also with products of intestinal microbiome, which may affect their metabolism and toxicity, including their mutagenicity. In particular, short chain fatty acids, such as acetate, propionate and butyrate belong among important groups of metabolites produced by the intestinal microbiome. Here, especially butyrate is known to be an important energy source for colon epithelial cells, while it simultaneously regulates cell proliferation, differentiation and cell death processes in both normal and tumor colon cells. However, its effects on the regulation of enzymes metabolizing and/or bioactivating carcinogens in the intestinal epithelium are relatively little known. In this dissertation, I present studies demonstrating that butyrate may significantly affect the expression and/or activities of enzymes involved in the metabolism of carcinogens, such as benzo[a]pyrene (BaP), in a number of cellular models derived from the colon epithelium. Butyrate enhances induction of the expression and/or activity of cytochrome P450 family 1 (CYP1) enzymes, which are regulated by AhR, which play a critical role in metabolism of BaP and related compounds. Therefore, it significantly affects formation of BaP metabolites, including production of ultimate reactive metabolites forming covalent DNA adducts. These effects of butyrate are linked to its inhibition of histone deacetylases (HDAC), which then control transcription of key AhR target genes, including CYP1A1. The effects of butyrate are not limited only to the CYP1 family of biotransformation enzymes but it may affect a wider range of enzymes involved in the biotransformation of xenobiotics and metabolism of carcinogens. This includes expression and/or activity of enzymes such as N acetyltransferases, NAD(P)H:quinone oxidoreductase 1 or UDP-glucuronosyltransferases, important enzymes involved in bioactivation or detoxification of dietary carcinogens. Our results demonstrate, for the first time, that butyrate, a common product of microbial fermentation of dietary fiber, may alter metabolism of toxic dietary contaminants, and thus modulate both their detoxification and/or formation of their genotoxic metabolites, within colon cells.
