In vitro models of type II pneumocytes and their applications in toxicity analyses of airborne pollutants

Abstract:

Pneumocyty typu II jsou součástí plicních sklípků, kde hrají významnou roli v regeneraci plicního epitelu, tvorbě surfaktantu a regulaci imunitních reakcí. Tyto buňky jsou přímo vystaveny jak působení patogenů, tak toxickému působení organických i anorganických polutantů obsažených ve znečištěném ovzduší. Částice polétavého prachu (PM) a s nimi asociované toxické látky mají negativní dopad na lidské zdraví i funkce buněk plicního epitelu. Z tohoto důvodu vzniká potřeba standardizovaných modelů pro studium působení toxických částic na dýchací soustavu. Cílem této bakalářské práce je shrnout dosavadní poznatky o modelech a možnostech kultivace pneumocytů typu II v in vitro podmínkách a poznatky o působení částic PM na různé typy modelů respiračního epitelu. Přestože je potvrzeno, že PM mají negativní dopad na funkce dýchací soustavy i kardiovaskulárního systému, o mechanismech jejich působení na lidské pneumocyty typu II je toho známo poměrně málo, což je dáno zejména nedostatkem vhodných buněčných modelů. Pokročilé metody kultivace plicních buněk a zejména příprava nových typů modelů lidských pneumocytů typu II mohou poskytnout nový typ informací o mechanismech působení PM a s nimi asociovaných organických polutantů, jako jsou polycyklické aromatické uhlovodíky, v tomto typu buněk.

Abstract:

Type II pneumocytes are part of the alveolus, where they play a major role in regeneration of alveolar epithelium, production of surfactant or regulation of immune reactions. They are directly exposed to pathogens or toxic action of both organic and inorganic pollutants found in ambient air. Both particulate matter (PM) and associated toxic compounds have a deleterious impact on human health, as well as on functions of pulmonary epithelium. Therefore, there is an increased need to develop standardized models for studies on impact of toxic particles on respiratory system. The aim of this bachelor thesis is to summarize current information about cellular models and methods for cultivation of type II pneumocytes under in vitro conditions, and the findings about the impact of PM on various respiratory epithelium cell models. Although it is known that PM have negative impact on respiratory system functions or cardiovascular system, due to limitations of cell models, we have very little information about the mechanisms of their toxic action in human type II pneumocytes. Advanced methods for cultivation of lung cells, and, in particular, preparation of new models of human type II pneumocytes may thus bring a novel type of information about the toxic modes of action of PM and associated organic pollutants, such as polycyclic aromatic hydrocarbons, in this type of cells.
