Analysis of resistance mechanisms of S. aureus strains to polyvalent phages using transposon mutagenesis

Abstract:

Staphylococcus aureus is an important human pathogen, that can cause serious diseases. Nowadays, their treatment is complicated by high incidence of strains resistant to broad spectrum of antibiotics. Therefore, alternative methods of treatment of bacterial infections are investigated. These also include the phage therapy. Although it has been successfully used for almost 100 years, this therapy has its shortcomings. One of them is bacterial resistance to phages. The mechanisms of phage resistance in S. aureus have been investigated a little so far. For that reason, this thesis has attempted to identify genes responsible for resistance to polyvalent phages in 5 strains of S. aureus. This was done by mutagenesis using the transposon Tn917. In this work, the method of electroporation of selected strains by plasmid vector pTV1ts carrying Tn917 was optimized. Using the successfully transformed strains, transposon mutants were prepared and subsequently subjected to screening for sensitivity to the polyvalent phage 812. However, the isolation of sensitive mutant with transposon inserted in a phage resistance gene was not successful. A mutant was identified which had lost resistance as the result of excision of prophage 53. The insertion site of Tn917 was localized into the gene lexA in the genome of this mutant.

Abstract:

Staphylococcus aureus je významný lidský patogen, který může způsobovat vážná onemocnění. V současnosti je jejich léčba komplikovaná vysokým výskytem kmenů rezistentním k široké škále antibiotik. Proto jsou zkoumány alternativní způsoby léčby bakteriálních infekcí, kterou je i fágová terapie. Přestože byla úspěšně používaná již téměř 100 let, má tato léčba své nedostatky. Jednou z nich je rezistence bakterií k fágům. Mechanismy fágové rezistence u S. aureus byly doposud málo prozkoumány. Tato práce se proto pokusila identifikovat geny odpovědné za rezistenci k polyvalentním fágům u 5 kmenů S. aureus. K tomu byla využita mutageneze pomocí transpozonu Tn917. V práci byla optimalizována metoda elektroporace vybraných kmenů plazmidovým vektorem pTV1ts, který nese Tn917. Z úspěšně transformovaných kmenů byly připraveny transpozonové mutanty, které byly následně podrobené skríningu na citlivost k polyvalentnímu fágu 812. Nepodařilo se však izolovat žádnou citlivou mutantu s transpozonem začleněným do genu fágové rezistence. Byla identifikována mutanta, která ztratila rezistenci v důsledku vyčlenění profága 53. V jejím genomu bylo inzerční místo Tn917 lokalizováno do genu lexA.
