A wormy world: exploring strongylid infections in wild primates following disease emergence in mountain gorillas

Abstract:

Strongylidní hlístice jsou celosvětově rozšířenou skupinou parazitů s medicínským i veterinárním významem ohrožující zdraví hostitele s případnými fatálními následky. Předpokládá se však, že u velkých volně žijících býložravců, jako jsou sloni, gorily a koňovití, jsou strongylidní hlístice běžnou součástí gastrointestinálního ekosystému, přičemž infekce jsou často asymptomatické. Přesto u ohrožených goril horských (Gorilla beringei beringei) bylz v posledních letech strongylidní infekce spojovány se zvýšeným výskytem gastrointestinálních onemocnění. U části populace goril ve Virunze byly během histopatologického vyšetření uhynulých jedinců nalezeny v žaludku léze typické pro strongylidní hlístice, zatímco bwindská populace trpí syndromem chronického chřadnutí, pro který je charakteristický úbytk hmotnosti a špatná tělesná kondice. Současné poznatky o strongylidních infekcích u volně žijících zvířat jsou omezené a výzkum gastrointestinálních parazitů většinou závisí na neinvazivním odběru trusu a mikroskopickém vyšetření. Tento přístup je však nedostatečný pro popis strongylidních hlístic, jež se vyskytují jako komplexní společenstva tvořena několika rody v rámci jednoho hostitele a vajíčka různých druhů nebo dokonce i rodů těchto hlístic jsou pod mikroskopem nerozlišitelná. Molekulární metody byly zatím aplikovány na jednotlivé parazity nebo byly využívány druhově specifické primery, kterými není možné popsat celá společenstva strongylidů. Nedávný vývoj v oblasti vysokokapacitního sekvenování umožnil vývoj rychlé a relativně levné metody pro popis celých společenstev strongylidů, včetně přesné identifikace, ačkoli použití tohoto přístupu u volně žijících zvířat je teprve v plenkách. V rámci své práce jsem za pomoci tradičních i moderních parazitologických technik popsala genetickou diverzitu společenstev strongylidů u volně žijících primátů a zároveň zkoumala vliv environmentálních a hostitelských faktorů na utváření těchto společenstev s následnou snahou lépe porozumět hrozbě, kterou mohou strongylidní hlístice představovat pro horské gorily. Konkrétně jsem nejdříve pomocí tradičních mikroskopických technik studovala vliv lidské činnosti a hostitelských faktorů na parazitární infekce včetně strongylidních hlístic u modelového hostitele paviána babuina (Papio cynocephalus). Dále jsem za pomocí ITS-2 metabarcodingu na MiSeq Illumina platformě získala poznatky o genetické diverzitě strongylidních hlístic u volně žijících goril nížinných (Gorilla gorilla gorilla) z několika lokalit za účelem porozumět variabilitě či konzistenci strongylidních společenstev napříč areálem rozšíření tohoto hostitele. Následně jsem popsala společenstva strongylidních hlístic u recentních a historických vzorků trusu goril horských a determinovala klíčové faktory, které je ovlivňují, za účelem objasnit výskyt klinických případů. Nakonec jsem popsala strongylidní společenstva u orangutanů bornejských (Pongo pygmaeus) a dalších sympatrických druhů asijských primátů, což umožnilo srovnání strongylidních infekcí u afrických a asijských lidoopů. Můj výzkum potvrdil výskyt komplexních společenstev skládajících se z několika rodů strongylidních hlístic u afrických lidoopů. Společenstva strongylidů u goril nížinných byla konzistentní napříč populacemi, přičemž dominantní rody Oesophagostomum a Necator tvořily jádro všech společenstev, která se lišila mezi jednotlivými populacemi pouze výskytem či relativní abundancí vzácnějších, satelitních taxonů. Naopak společenstva strongylidních hlístic u horských goril nebyla geograficky konzistentní a zásadně se lišila od společenstev u goril nížinných.

Abstract:

Heterogenní strongylidní společenstva ovlivněná vnějšími faktory byla zaznamenána dokonce i v rámci jedné populace goril ve Virunze. V rámci této populace byly u skupin goril z oblasti s nevyšším výskytem gastritid zaznamenány vysoké absolutní abundance patogenních žaludečních strongylidních hlístic rodu Hyostrongylus. Společenstva strongylidů s dominantními patogenními druhy se historicky u goril nevyskytovala, objevila se až v posledních dvou desetiletích. Výsledky tak objasnily recentní výskyt gastritid u goril horských. Překvapivě u studovaných asijských primátů včetně orangutanů bornejských byla zaznamenána geograficky konzistentní společenstva tvořena pouze jedním dominantním druhem, kterým je Oesophagostomum aculeatum. Můj výzkum výrazně posunul naše znalosti o strongylidních infekcích u volně žijících primátů a potvrdil, že změny v epidemiologii parazitů je třeba považovat za hrozbu pro ochranu ohrožených zvířat, ale zároveň i poukázal na mezery ve znalostech týkajících se strongylidních společenstev u sympatrických druhů.

Abstract:

Strongylid nematodes are a globally distributed parasitic group with both medical and veterinary importance, able to pose a serious threat to host health and even be fatal. However, strongylids are believed to be a usual part of the gastrointestinal ecosystem of wild, large-bodied herbivores, such as elephants, gorillas, and equids, with infections often asymptomatic. Despite this, in recent years strongylid infections have been associated with increased gastrointestinal disease occurrence within endangered mountain gorillas (Gorilla beringei beringei). In part of the Virunga gorilla population, clinical symptoms result from lesions typical of gastric strongylid infections, identified during histopathological examination of deceased individuals, while chronic wasting syndrome, characterised by weight loss and poor body condition, is reported amongst the Bwindi gorilla population. Current knowledge of strongylid infections in wildlife is limited, with investigation of gastrointestinal parasites often depending on non-invasive collection of faecal material and microscopic analysis. Such an approach is insufficient for describing strongylid nematodes, which occur as complex, multi-genera communities within a single host and eggs of different species, or even genera, are indistinguishable under the microscope. Previous applications of molecular methods are centred upon use of single parasite specimens or species-specific primers, unable to describe the entire strongylid community. Recent developments in high-throughput sequencing have enabled quick, relatively low-cost, methodologies for describing entire strongylid communities, including strain-level identification, though application to wildlife is in its infancy. This thesis describes the genetic diversity of wild primate strongylid communities while exploring the environmental and host variables which may shape them, subsequently seeking to better understand the conservation threat strongylids pose to mountain gorillas by combining both traditional and modern parasitological techniques. Specifically, this thesis first investigates the association of human disturbance and host factors with parasite infections, including strongylid nematodes, through traditional microscopic approaches, utilising yellow baboons (Papio cynocephalus) as a model. Secondly, through application of ITS-2 metabarcoding at the MiSeq Illumina platform, this thesis establishes baseline data on the genetic diversity of strongylid infections in wild gorillas, surveying western lowland gorillas (Gorilla gorilla gorilla) across multiple localities to gain insight into the variability or consistency of strongylid communities across geographical range. This thesis then describes the strongylid communities of mountain gorillas, both recently and historically, investigating the determinants of strongylid communities which can explain clinical case emergence. Lastly, the strongylid communities of Bornean orangutans (Pongo pygmaeus), and their sympatric primate species, were described, providing a comparison between African and Asian great apes. My research confirmed occurrence of complex, multi-genera strongylid communities in African apes. A general consistency of strongylid communities was demonstrated in western lowland gorillas, with dominant Oesophagostomum and Necator forming core genera prevalent within all surveyed populations and only occurrence or relative abundance of rarer, satellite taxa varying. Crucially, strongylid communities of mountain gorillas differed from those of lowland gorillas and lacked geographic consistency, with heterogenous communities, affected by extrinsic factors, revealed within the Virunga population. In this population, high absolute abundances of strongylids, from the pathogenic gastric genus Hyostrongylus, were identified in the areas where gastritis cases were most reported. ades. These results thus explain recent emergence of gastritis in mountain gorillas. Surprisingly, studied Asian primates harboured single-species strongylid communities dominated by Oesophagostomum aculeatum, yet geographical consistency remained. This research greatly improves our knowledge of strongylid infections in wild primates and highlights how changes in parasite epidemiology must be considered as a conservation threat amongst endangered animals, yet also demonstrates knowledge gaps pertaining to the strongylid communities of sympatric species.
