Solution- and Solid-State NMR Parameters: Applications in Structural Chemistry

Abstract:

NMR spektroskopie je nedílnou součástí moderní chemie. Zatímco metodologie pro studium roztoků je využívána rutinně, metody NMR pevné fáze se začaly dostávat do širšího povědomí teprve v průběhu minulých dvaceti let, kdy prošly zásadním rozvojem. Již tento fakt ukazuje na to, že pevná fáze je pro NMR spektroskopii o dost složitějším systémem než fáze kapalná. Proto jsou v úvodní kapitole této práce stručně shrnuty základní NMR parametry obou těchto fází s cílem poukázat na jejich rozdíly. V dalších kapitolách jsou pak tyto NMR parametry korelovány s různými aspekty struktury molekul a krystalové struktury. Stěžejní část práce se zabývá využitím tenzoru chemického posunu ve strukturní analýze. Nejprve jsou v krátkosti popsány experimentální a výpočetní metody používané ke zjištění tohoto NMR parametru a následně jsou diskutovány jeho vztahy ke struktuře molekul a krystalů. Důraz je zde kladen zejména na vliv substituce, tautomerie a mezimolekulárních interakcí na tenzor chemického posunu, neboť tato část práce přináší významné nové poznatky. Druhá kapitola této práce je zaměřena na přímou jadernou spin-spinovou interakci a její využití k určování mezijaderných vzdáleností a charakteristik molekulárního pohybu. Tato kapitola v krátkosti vysvětluje princip nové metody měření této interakce založené na analýze Hartmann-Hahnových profilů získaných pomocí LGCP experimentu (cross-polarizace za současné aplikace Lee-Goldburgova decouplingu). V poslední části práce je pozornost věnována nepřímé jaderné spin-spinové interakci a novým pozorováním v oblasti vztahů mezi elektronovou strukturou molekulových fragmentů, které tuto interakci zprostředkovávají v molekule purinu, a silou této interakce vyjádřenou příslušnou interakční konstantou. Jako celek práce dokumentuje široké možnosti využití NMR spektroskopie kapalného i pevného stavu ve strukturní chemii.

Abstract:

NMR spectroscopy is an integral part of modern chemistry. While the solution-state NMR methods are well established, the intensive development in the field of solid-state NMR is a matter of last two decades. It points to the fact that the solid phase represents for NMR spectroscopy much more complex system than the liquid phase. In this thesis, the differences between the main solution-state and solid-state NMR parameters are briefly summarized in an introductory chapter. In the next chapters, the introduced NMR parameters are related to various aspects of molecular and crystal structure. The main part of the thesis focuses on the application of chemical shift tensor data in structural analysis. Firstly, the experimental and computational methods used to characterize the chemical shift tensors are briefly described. Further, the different kinds of structural information provided by the data are discussed. In this chapter, the focus is mainly on the effects of substitution, tautomerism, and intermolecular interactions on the chemical shift tensor data, where several significant new findings are introduced. Second part of the thesis deals with direct nuclear spin-spin coupling and its use for the study of internuclear distances and motional parameters by using Lee-Goldburg cross-polarization (LGCP) technique. This chapter serves as an introduction to a newly developed methodology based on analysis of Hartmann-Hahn matching profiles. Last part of the thesis is focused on the indirect nuclear spin-spin coupling and the newly observed trends in the relation between the electronic structure of fragments mediating the coupling in the molecule of purine and the magnitude of the observed coupling. The thesis as a whole tries to present the variety of structural problems in chemistry, which can be solved by analyzing the solution- and solid-state NMR parameters.
