Nitisinone, herbicide and drug and its effect on parasitic and non-parasitic nematodes.

Abstract:

Diplomová práce se zabývala studiem účinku nitisinonu (NTBC), inhibitoru enzymu 4-hydroxyfenylpyruvát dioxygenázy (HPPD), na vybrané zástupce hlístic s využitím fenotypových experimentů, molekulárně-biochemických metod a bioinformatické analýzy. Fenotypové testy provedené na modelovém organismu Caenorhabditis elegans (n = 13) prokázaly silný synergický účinek NTBC v přítomnosti exogenního L-tyrosinu. Zatímco samostatné podání látek mělo na vitalitu hlístic minimální vliv, jejich kombinace vedla k signifikantnímu poklesu reprodukce o 44,2 % a vysoké akutní mortalitě dosahující 61,5 %. Tento jev potvrzuje, že k fatálnímu poškození organismu nevede samotná blokáda enzymu, ale až následná akumulace toxických meziproduktů. Předběžné testy u fytoparazita Meloidogyne hapla odhalily extrémní fytotoxicitu NTBC vůči hostitelským rostlinám již ve fázi klíčivosti, což znemožnilo vytvoření hostitelského systému pro fenotypové studie a poukázalo na limity praktického využití látky v ochraně rostlin. Molekulárně-biochemická část práce byla zaměřena na rekombinantní expresi HPPD u druhů M. hapla a C. elegans v systému E. coli. Kolorimetrická esej nevykázala u parazitárních proteinů očekávanou pigmentaci. Ve spolupráci s Biologickým centrem AV ČR bylo pomocí metody Western Blot zjištěno, že sledované enzymy tvoří v bakteriálním hostiteli neaktivní inkluzní tělíska, což vysvětluje absenci jejich enzymatické aktivity. Zjištění poukazují na zásadní limity standardních expresních protokolů pro parazitární proteiny a definují nutnost budoucí optimalizace experimentálních podmínek při studiu rekombinantních proteinů u parazitických hlístic. Bioinformatická analýza 11 sekvencí potvrdila vysokou konzervaci aktivního místa enzymu napříč studovanými řády hlístic i zooparazitem Haemonchus contortus. Identifikována byla absolutní shoda v klíčové katalytické triádě (His183, His266 a Glu349) a vysoká míra konzervace u reziduí určujících vazebnou afinitu inhibitoru (Tyr221 a Leu224), což podporuje hypotézu o univerzální zranitelnosti tyrosinové dráhy u nematod. Získané výsledky naznačují, že HPPD představuje perspektivní cíl pro vývoj anthelmintik, jehož účinnost je však úzce spjata s dostupností L-tyrosinu. Práce zároveň upozorňuje na environmentální rizika pro volně žijící hlístice.

Abstract:

The diploma thesis focused on the study of the effect of nitisinone (NTBC), an inhibitor of the enzyme 4-hydroxyphenylpyruvate dioxygenase (HPPD), on selected nematode species using phenotypic experiments, molecular-biochemical methods, and bioinformatic analysis. Phenotypic assays conducted on the model organism Caenorhabditis elegans (n = 13) demonstrated a strong synergistic effect of NTBC and exogenous L-tyrosine. While individual treatments had only a minimal impact on nematode viability, their combination resulted in a significant reduction in reproduction by 44.2 % and a high acute mortality rate reaching 61.5 %. These findings indicate that fatal effects are not caused by enzyme inhibition alone, but rather by the subsequent accumulation of toxic metabolic intermediates. Preliminary experiments with the phytoparasitic nematode Meloidogyne hapla revealed extreme phytotoxicity of NTBC toward host plants already at the germination stage, which prevented the establishment of a host system for phenotypic assays and highlighted limitations in its practical application in plant protection. The molecular-biochemical part of the study focused on the recombinant expression of HPPD from M. hapla and C. elegans in an E. coli system. The colorimetric assay did not show the expected pigment formation for the parasitic proteins. In collaboration with the Biology Centre of the Czech Academy of Sciences, Western blot analysis revealed that the studied enzymes formed inactive inclusion bodies in the bacterial host, explaining the absence of detectable enzymatic activity. These findings highlight significant limitations of standard expression protocols for parasitic proteins and emphasize the need for further optimization of experimental conditions when studying recombinant proteins in parasitic nematodes. Bioinformatic analysis of 11 protein sequences confirmed a high degree of conservation of the enzyme’s active site across the studied nematode orders as well as in the zooparasite Haemonchus contortus. Complete conservation was identified in the key catalytic triad (His183, His266, and Glu349), along with a high level of conservation in residues determining inhibitor binding affinity (Tyr221 and Leu224), supporting the hypothesis of a broadly conserved vulnerability of the tyrosine degradation pathway in nematodes. The results suggest that HPPD represents a promising target for anthelmintic development; however, its effectiveness appears to be closely linked to the availability of L-tyrosine. At the same time, the study highlights potential environmental risks for free-living nematodes.
