Identification of the onset and intensity of drought stress in maize

Abstract:

Sucho patří k hlavním abiotickým faktorům limitujícím produktivitu kukuřice. Dopady stresu z nedostatku vody na transpiraci a produkci biomasy dosahují různé úrovně v závislosti na termínu jeho nástupu, síle a době trvání. Cílem této práce bylo posoudit vliv stupňované intenzity stresu suchem na úroveň transpirace, biometrické a výnosové parametry, expresi genů pro dehydriny a kořenový systém rostlin kukuřice, kvantifikovat míru změny těchto procesů a vlastností a identifikovat nástup stresu v nádobovém pokusu v částečně řízených podmínkách. Stres suchem byl navozen v době metání lat na úrovni 42 %, 28 % a 25 % využitelné vodní kapacity půdy (VVK) a výsledky byly srovnávány s kontrolní variantou s optimální úrovní dostupnosti půdní vláhy. Výnos biomasy a zrna reagoval na nedostupnost vody méně výrazně než intenzita transpirace a exprese genů. Byl zjištěn významný vliv stresu suchem na úrovni 25–28 % VVK na intenzitu transpirace, úroveň exprese genů ZmDHN1 a ZmDHN2 a vlastnosti kořenového systému. Vliv změny úrovně závlahy na intenzitu transpirace byl pomocí metody měření sap flow detekován po 14 dnech podobně jako exprese genů pro dehydriny. S intenzitou a délkou trvání stresu se snižovala závislost transpirace na evapotranspiračních požadavcích atmosféry, tj. globální radiaci a sytostním doplňku. Efektivita využití vody (WUE) se za stresu suchem zvýšila. Byly zjištěny genotypové rozdíly pro intenzitu transpirace, expresi genů a vlastnosti kořenového systému v závislosti na síle stresu. Nižší intenzitu transpirace, vyšší expresi genů pro dehydriny a vyšší hodnoty délky a plochy kořenového systému v povrchové vrstvě půdy za silného stresu suchem vykazovaly rostliny stres-senzitivního genotypu.

Abstract:

Drought is one of the main abiotic factors limiting maize productivity. The effect of water stress on transpiration and biomass production reach different levels depending on the date of its onset, severity and duration. The aim of this work was to assess the effect of a graded drought stress intensity on the transpiration rate, biometric and yield parameters, dehydrin gene expression and root system of maize plants, quantify the rate of changes in these processes and characteristics and identify the onset of stress in the pot experiment under partially regulated conditions. Drought stress was induced during the tasseling stage at the levels of 42%, 28% and 25% of available water holding capacity (AWHC) and the results were compared with a control treatment at an optimal level of AWHC. Biomass and grain yield responded to water scarcity less significantly than the transpiration and gene expression. A significant effect of drought stress on the transpiration, the level of expression of ZmDHN1 and ZmDHN2 genes and the root system characteristics was observed at the level of 25–28% AWHC. The effect of changes in irrigation level on transpiration rate was reflected by the method of sap flow measuring after two weeks, as well as the dehydrin genes expression. The relationship of transpiration and the evapotranspiration demands, i.e. global radiation and vapour pressure deficit, weakened with the increasing intensity and duration of stress. Water use efficiency (WUE) increased as a result of drought stress. Genotypic differences were observed in transpiration rate, gene expression and root system parameters depending on the severity of stress. Lower transpiration rate, higher expression of dehydrin genes and higher values of length and surface area of the root system in the shallow soil layer under severe drought stress were shown by plants of a stress-sensitive genotype.
