Cloning of structural capsid genes of phage 812 and theit expression for 3D structural analysis

Abstract:

Bacteriophage 812 represents potential candidate for the treatment of the infection caused by antibotic-resistant strains of Staphylococcus aureus. In this work, three of its strucural proteins had been studied, putative hydrolase TmpE, portal protein Prt and morphogenic protein of the baseplate TmpF. The genes encoding above-mentioned proteins were successfully cloned into the expression vector, fused to C-terminal histidine tag. Subsequently, they were over-expressed in Escherichia coli and the products were purified by metal-affinity and size-exclusion chromatography. The results include yielded crystals of the putative hydrolase, suitable for further optimization, functional complexes of portal protein usable for single-particle reconstruction by cryo-electron microscopy and purified protein TmpF, suitable for functional assays.

Abstract:

Bakteriofág 812 představuje potencionálního kandidáta pro léčbu infekcí způsobených k antibiotikám-rezistentními kmeny Staphylococcus aureus. V této práci byly studované jeho tři strukturní proteiny, domnělá hydroláza TmpE, portálový protein Prt a morfogenetický protein bazální destičky TmpF. Odpovídajíci geny byly naklonovány do expresního vektoru za vzniku konstruktů nesoucích C-koncovou histidinovou kotvu. Následně byly geny exprimovány v Escherichia coli a purifikovány metalo-afinitní a gelovou permeační chromatografii. Výsledkem jsou krystaly domnělé hydrolázy s vhodnými vlastnostmi pro další optimalizaci, funkční komplexy portálového proteinu, použitelné pro rekonstrukci obrazu částice kryo-elektronovou mikroskopií a přečištěný protein TmpF, vhodný pro funkční studie.
