Photochemical release of carbon monoxide from alkynyl flavonols

Abstract:

Malé organické molekuly, které jsou schopné uvolnit po ozáření oxid uhelnatý (fotoCORMy), se ukazují jako slibná cesta terapie nádorových onemocnění. Přestože již byla popsána řada takových sloučenin, většina z nich stále postrádá některé vyžadované vlastnosti, jako je například rozpustnost ve vodě nebo dostatečně účinné uvolňování CO. Tato práce se zabývá flavonoly jako slibnými kandidáty na fotoCORMy. V literární části je popsán mechanismus fotoindukovaného uvolňování CO z těchto látek a vliv různých substituentů na tento proces. Experimentální část pak pojednává o syntéze nových derivátů flavonolů s ethynylovou funkční skupinou, jejich spektroskopickou charakterizací a určení chemických a kvantových výtěžků uvolněného oxidu uhelnatého. Díky přítomnosti trojné vazby došlo k bathochromnímu posunu ve spektrech připravených derivátů ve srovnání s nesubstituovanými flavonoly a navíc jedna z připravených látek vykazovala vysoké výtěžky a účinné uvolňovaní CO. Trojná vazba by tudíž mohla sloužit jako užitečný strukturní motiv ve vývoji dalších fotoCORMů.

Abstract:

Simple organic molecules, which can release carbon monoxide upon irradiation with visible light (photoCORMs), have emerged as a promising strategy for cancer treatment. Although several promising compounds have already been described, they still have not met all the required conditions, e.g., sufficient solubility in water or efficiency of the CO release. In this thesis, flavonols are presented as auspicious photoCORM candidates. Mechanism of the CO release from these compounds and the effect of various substituents on this process are described in the literature part, whereas the experimental part deals with the synthesis of novel flavonol derivatives bearing an ethynyl group and their spectroscopic characterization, as well as the determination of chemical and quantum yields of released carbon monoxide. The ethynyl group provides bathochromically shifted absorption spectra of the synthesized compounds compared to unsubstituted flavonols. Furthermore, one of the prepared compounds showed an efficient release of high amounts of CO. Hence, this substitution could serve as a useful structural motif in the further photoCORM development.
