Apoptosis as a terapeutic target in cancer treatment

Abstract:

Apoptóza je programovaná buněčná smrt, která, díky činnosti specifických proteáz a pro-apoptotických a anti-apoptotických proteinů, udržuje v organizmu homeostázu. Narušení rovnováhy mezi těmito proteiny má za následek nadměrné odumírání buněk nebo vznik rezistence k buněčné smrti. Nádorové bujení často vzniká jako následek rezistence buněk podstoupit apoptózu, a proto je porucha tohoto mechanizmu buněčné smrti jedním z hlavních znaků rakoviny. U různých typů nádorů jsou dysregulovány jednotlivé proteiny apoptotických drah, což přispívá k procesu nádorové transformace i k rezistenci k terapii. Jednotliví hráči apoptotických drah jsou tedy vhodnými terči protinádorové terapie. Cílem bakalářské práce je popis mechanizmu apoptózy, proteinů, které se tohoto procesu účastní a sloučenin, které cíleně ovlivňují tyto proteiny a jsou v současné době ve vývoji, v klinických testech, nebo jsou již používány k léčbě nádorových onemocnění.

Abstract:

Apoptosis, a form of programmed cell death, maintains homeostasis in the organism through the activity of specific proteases, proapoptotic and nonapoptotic proteins. Disharmony between these proteins causes excessive dying of cells or developement of resistence towards cell death. Tumor cell growth often unsues when cells develop resistence to apoptosis, which makes the disorder of this mechanism one of the main symptoms of cancer. In different types of tumors the different apoptotic proteins are dysregulated, which leads to tumor cell transformation and resistance to therapy. Therefore, the individual players of apoptotic pahtways are appropriate tumor therapy targets. This bachelor´s thesis aims to describe the mechanism of apoptosis, the proteins effective during the proces and compounds which influence these proteins and are currently being developer, clinically tested, or have already been used to cure cancer.
