Transcription factors analyses of anthocyanins biosynthetic pathway

Abstract:

Antokyany jsou přírodní barviva hojně využívaná v potravinářství mimo jiné i pro svoje antioxidační účinky. Biosyntetická dráha antokyanů a její transkripční faktory jsou často studované u mnoha kulturních druhů. Při pochopení biosyntézy a faktorů, které ji ovlivňují, by genotypy pšenice seté (Triticum aestivum L.) produkující antokyany v obilkách mohly být vylepšovány šlechtěním a využity pro výrobu funkčních potravin. Hlavním cílem této práce bylo označit genomové oblasti zajímavé z hlediska biosyntézy antokyanů, analýza sekvence genu TaMYC1, který svojí expresí koreluje s expresí strukturních genů, u vybraných genotypů a ověření schopnosti vazby na promotor genu F3H. Tyto poznatky mohou přispět ke zlepšení šlechtitelských přístupů při selekci na purpurově nebo modře zabarvené obilky.

Abstract:

Anthocyanins are natural pigments abundantly used in the food industry because of their antioxidative effects. Their biosynthetic pathway and its transcription factors are frequently studied in many species. Better understanding of the biosynthesis of anthocyanins and the factors by which it is influenced would help in breeding of application-improved genotypes of anthocyanins-producing bread wheat (Triticum aestivum L.). Subsequently, they could be utilized for the production of functional food. The aim of this thesis was to map the genome sequences linked to anthocyanins biosynthesis. Furthermore, in selected genotypes, the thesis analyzes the sequence of TaMYC1 gene, whose expression correlates with the expression of structural genes, and tests its ability to bind the promotor of the F3H gene. Yielded findings will contribute to the improvement of breeding approaches in the selection of purple or blue grains.
