Abstract:

Fyzikální dormance semen rostlin z čeledi bobovitých byla v průběhu domestikačního procesu pozměněna a výsledkem tohoto procesu jsou nedormantní semena kulturních plodin s osemením propustným pro vodu, která zajišťují uniformní vyklíčení. Oproti tomu semena jejich planých předků zůstávají dormantní a fyzikální dormance semen je dána převážně chemickým složením a tloušťkou osemení. Byla provedena kombinovaná studie dormance semen pro kontrastní rodičovské genotypy cizrny a populace rekombinantních inbredních linií (RILs). Testování klíčivosti semen odhalilo rozdíly mezi rodičovskými genotypy, kdy kulturní cizrna vyklíčila během 24 h, zatímco planá cizrna dosáhla 100% klíčivosti po 20 dnech pozorování. Anatomická skladba osemení odhalila rozdíl v délce makrosklereid rodičovských genotypů cizrny a populace RILs. Histochemické barvení osemení odhalilo rozdíly mezi osemením rodičovských genotypů cizrny a RILs a to zejména při detekci sloučenin lipidů. Analýza metabolitů v osemení odhalila rozdíly v obsahu fenolických látek (např. kyselina gallová) a flavonoidů (např. myricetin) detekovaných u rodičovských genotypů a RILs. Genetické mapování a analýza genové exprese osemení cizrna měla za cíl užší lokalizaci či přímo identifikaci genů zodpovědných za ztrátu dormance semen v průběhu domestikace cizrny. Genetické restrikční mapování odhalilo rozdíly převážně mezi rodičovskými genotypy. Analýza genové exprese vybraných diferenciálně exprimovaných genů osemení stanovila rozdíly exprese v daných časových intervalech mezi rodičovskými genotypy cizrny a vybranými liniemi.

Abstract:

The physical dormancy of the legume seeds has been altered during the domestication process, resulting in non-dormant crop seeds with seed coats permeable for water that provides fast and uniform germination. In contrast, the dormant seeds of wild ancestors remain dormant and the physical dormancy is mediated by chemical composition and seed coat thickness. A combined analyses of seed dormancy was performed for contrasting parental genotypes of chickpea and recombinant inbred lines (RILs) population. Germination testing revealed the differences between parental genotypes, with cultivated chickpea germinating within 24 h, while wild chickpea reached 100% germination after 20 days. Anatomical composition of the seed coats revealed a significant difference in lenght of the macrosclereids of parental genotypes and RILs population. Histochemical staining revealed differences between seed coats of parental genotypes and RILs especially in the detection of lipid compounds. The analysis of metabolic content of the seed coats revealed differences in the content of phenolics (e.g. gallic acid) and flavonoids (e.g. myricetin) detected in parental genotypes and RILs. Genetic mapping and gene expression analyses were performer with the chickpeas seed coats with the aim of narrower localization or direct identification of genes responsible for the seed dormancy loss during domestication process of chickpea. Genetic restriction mapping revealed differences between parental genotypes. Gene expression analyses of selected differentially expressed genes of the seed coats determined expression differences in gived time intervals between parental genotypes and selected RILs.
