Impact of viral infection on the immune gene expression in fish

Abstract:

Imunitní systém organizmů představuje silnou obranu proti rozličným patogenům. Tato bakalářská práce pojednává o imunitní odpovědi ryb na virovou infekci. Práce shrnuje expresi imunitních genů zapojených v antivirové odpovědi, a poskytuje přehled deseti ekonomicky důležitých virů způsobujících vážné ztráty v chovu na rybích farmách. Jedním ze způsobů studia antivirové imunitní odpovědi organizmů je analýza genové exprese. Metoda qPCR je považována za nejpřesnější a je proto nejběžněji používána pro zmíněnou analýzu. Výsledky analýz exprese genů zapojených do antivirové imunitní odpovědi získané prostřednictvím metody qPCR tvoří jádro této práce. Pomocí qPCR byla v praktické části práce realizována analýza exprese genů TLR3 a IL-1β v osmi tkáních hybridů Carassius auratus × Cyprinus carpio a jedinců C. auratus. Rozdíly v expresi genů TLR3 a IL-1β byly nalezeny mezi neinfikovanými hybridy a hybridy infikovanými virem jarné virémie kaprů (SVCV), a mezi neinfikovanými hybridy a neinfikovanými jedinci C. carassius. Pochopení genové exprese během antivirové imunitní odpovědi může přispět k účinnější prevenci virových ochoření, a tím pozdvihnout úroveň chovu na rybích farmách a zlepšit zdravotní stav volně žijících ryb.

Abstract:

The immune system of an organism forms a powerful defence against diverse pathogens. This thesis is focused on fish immune response to viral infection. The work summarizes the expression of immune related genes involved in antiviral response, and provides an overview of ten economically important viruses of fish causing significant losses in fish farms. One of possible ways how to study antiviral immune response of organisms is gene expression analysis. The qPCR method is considered to be the most precise and, therefore, most common method used in gene expression analysis. Results of studies on antiviral immune-related genes expression applying qPCR formes a core of the thesis. Using qPCR approach, the practical section involves the gene expression analysis of TLR3 and IL-1β genes in eight distinct tissues of Carassius auratus × Cyprinus carpio hybrids and C. auratus individuals. Differences in TLR3 and IL-1β genes expression patterns were reported between the uninfected hybrids and hybrids infected with spring viraemia of carp virus (SVCV), and between the uninfected hybrids and uninfected C. carassius fish. Understanding of expression patterns in fish antiviral immune response might contribute to efficient prevention of viral diseases, and subsequently to enhancement of fish farming and health status of free-living fish.
