The role of chromatin in telemere length regulation in arabidopsis

Abstract:

Doktorská práce se zaměřením na studium vlivu chromatinu na stabilitu telomer. Současný biologický výzkum přináší mnoho nových poznatků nezbytných pro porozumění biologie telomer. Jednotlivá studia využívají výhod různých modelových organismů k charakterizaci konkrétních detailů. Nejnovější metody molekulární biologie zahrnují celo-genomové analýzy s vysokým rozlišením, studium regulace na úrovni malých RNA molekul, chromatinové imunodetekční techniky a další. S využitím výsledků současného výzkumu a s použitím dat získaných moderními technologiemi jsme se rozhodli prozkoumat strukturu chromatinu na terminálních chromozomových doménách u Arabidopsis thaliana a pokusit se tak přispět k pochopení role chromatinu při regulaci telomer. Velkou pozornost jsme věnovali transktipci telomer a siRNA molekulám, které jsou odvozené z DNA sekvencí obsahujících telomerové repetice. Rovněž jsme se zaměřili na studium DNA a histonové metylace subtelomer a telomer. Analyzovali jsme expresní profily genů, které se u Arabidopsis nacházejí v těsné blízkosti telomer a porovnali jsme získaná data s expresními profily těchto genů u známých mutantů, charakteristických výraznými změnami v délce telomer. Nakonec jsme se zaměřili na regulační roli siRNA na řízení metylace telomer a tím možnému ovlivnění transkripce částí DNA, které obsahující telomerové repetice.

Abstract:

This PhD thesis is focused at the telomere biology in connection to the chromatin structure. Research results on the field of the current biology uncovered many important novel findings in understanding of the telomeres. Individual studies use the advantages of various model organisms to describe particular details. Sophisticated methods of molecular biology include high resolution genome analyses, small RNA molecules studies, chromatin immunostaining techniques and many others. Taking the benefit of these facts we decided to examine the chromatin structure at the chromosomal termini in Arabidopsis in details and explain its regulatory function at telomeres. In particular, we focused at transcription of telomeres and siRNA which are derived from telomere-repeat-containing sequences of DNA. We also studied the methylation of terminal chromosomal domains – subtelomeres and telomeres. In details, we analyzed the expression of subtelomere genes and compared these results to expression profiles of telomere length-mutants. Finally we focused at possible regulation of telomeric chromatin and transcription by mechanism of RNA directed DNA methylation.
