Characterisation of TERT genes in plants of the Solanaceae family

Abstract:

Nedávné výsledky telomerového výzkumu ukázaly existenci několika paralogů genu pro telomerovou reverzní transkriptázu - TERT v rostlinném genomu amfidiploidního druhu Nicotiana tabacum a existenci odpovídajících forem těchto genů v jeho rodičovských druzích, N. sylvestris a N. tomentosiformis. Cílem této DP bylo charakterizovat sekvence genů TERT nebo jejich fragmenty u dalších zástupců čeledi Solanaceae, zjistit, zda přítomnost více kopií a variant genu TERT je v Solanaceae ojedinělý nebo běžný jev a provést fylogenetickou analýzu jejich příbuznosti a porovnat získané výsledky s dostupnými údaji o fylogenezi v rámci čeledi Solanaceae. Získané výsledky ukazují, že existence více paralogů TERT, zjištěná dříve u N. tabacum a N. sylvestris, není v rámci rodu Nicotiana ojedinělá. Přítomnost dvou variant byla zjištěna také u N. rustica a N. clevelandii, zatímco v případě N. noctiflora a N. repanda byly zjištěny dokonce tři varianty TERT. Po dvou variantách TERT bylo nalezeno rovněž u několika zástupců čeledi Solanaceae mimo rod Nicotiana - Juanolla aurantica, Brunfelsia calycina a Solanum elipticum. V případě dvou variant TERT nalezených v alotetraploidní N. rustica byl na základě sekvenčních dat zjištěn původ těchto variant z rodičovských genomů, N. paniculata a N. undulata. Tzv. D-varianta TERT, která byla dříve zjištěna u N. tabacum a N. sylvestris a je považována za pseudogen s možnou regulační funkcí, byla nalezena rovněž u několika dalších zástupců rodu Nicotiana a detailně charakterizována v případě N. noctiflora. Na základě získaných údajů byl zmapován její předpokládaný vznik v rámci fylogeneze rodu Nicotiana. Získané sekvence vybraného úseku TERT u zástupců Solanaceae byla využita k fylogenetickým analýzám, které ukázaly, že fylogeneze TERT odpovídá dostupným fylogenetickým údajům z jiných zdrojů.

Abstract:

Recent results have shown the existence of several paralogs of the gene for telomere reverse transcriptase - TERT in the plant genome of amphidiploid species Nicotiana tabacum, and the existence of appropriate forms of these genes in the parental species, N. sylvestris and N. tomentosiformis. The aim of this thesis was to characterize the TERT gene sequences, or fragments in other members of the family Solanaceae, to determine if the presence of multiple copies of a TERT gene variants in the Solanaceae is rare or common phenomenon and perform phylogenetic analysis of their relatives and compare the obtained results with available data about the phylogeny within the family Solanaceae. The results obtained show that the existence of multiple TERT paralogs, which was found previously in N. tabacum and N. sylvestris, is not unique within Nicotiana genus. Presence of two variants was observed also in N. rustica and N. clevelandii, while three variants were found in N. noctiflora a N. repanda. In several Solanaceae species outside of Nicotiana genus (Juanolla aurantica, Brunfelsia calycina and Solanum elipticum), two TERT variants were also observed. In the case of two TERT variants found in allotetraploid N. rustica, their origin from genomes of its progenitor species - N. paniculata a N. undulata was established based on sequencing data. The TERT variant termed as the D-variant, that has been previously found in N. tabacum a N. sylvestris, and considered as a pseudogene with a regulatory function, was also detected in several other Nicotiana species and was characterized in detail in N. noctiflora. Based on the data obtained, its origin during phylogeneis of the Nicotiana genus was mapped. The obtained sequences of the selected TERT segment in Solanaceae representatives were used for fylogenetic analyses which showed agreement of TERT molecular evolution with the available phylogenetic data from independent resources.
