Chemopreventive effects of phytochemicals – in vitro evaluation

Abstract:

Tato diplomová práce se zabývá chemopreventivními a protinádorovými účinky fytochemikálií, které byly posuzovány in vitro metodami. Chemopreventivní vlastnosti byly hodnoceny dle účinků těchto látek na mezibuněčnou komunikaci zprostředkovanou mezerovými spoji (GJIC) potkaních jaterních buněk WB F344. Protinádorové účinky dvou vybraných fytochemikálií byly navíc posuzovány dle jejich vlivu na buněčnou proliferaci, indukci apoptózy a buněčný cyklus u WB F344 buněčné linie trasfekované H-ras onkogenem. Pro tyto účely bylo vybráno devět fytochemikálií nebo jim příbuzných látek: dialyl sulfid, kyselina skořicová, kurkumin, emodin, indol-3-karbinol, metformin, quercetin, silibinin a tymochinon. U těchto látek byl posuzován vliv na chemicky indukovanou inhibici mezibuněčné komunikace. Inhibice GJIC byla zajištěna pomocí šesti známých inhibitorů a nádorových promotorů: 1,1,1-trichlor-2,2-bis(4-chlorfenyl)ethanu (DDT), fluoranthenu, lindanu, pentachlorofenolu (PCP), kyseliny perfluorooktanové (PFOA) a 12 tetradekanoyl 13 forbolesteru (TPA). Testy ukázaly, že všechny z uvedených fytochemikálií jsou schopny preventivně působit proti inhibici alespoň jednoho inhibitoru. Tato skutečnost potvrzuje, že vybrané fytochemikálie mají potenciálně chemopreventivní účinky. Dále bylo prokázáno, že metformin a silibinin jsou schopny obnovit GJIC, indukovat apoptózu a ovlivňovat buněčný cyklus u tumorigenní linie WB F344-ras. Selektivní účinky metforminu a silibininu na proliferaci tumorigenních buněk nebyly prokázány. Všechny tyto efekty nasvědčují, že by se mohlo jednat o látky, které jsou schopny účinně působit na vývoj nádorových onemocnění.

Abstract:

This diploma thesis focused on chemopreventive and anticancer effects of phytochemicals and their analogues, which were evaluated using in vitro methods. Gap junctional intercellular communication (GJIC) in rat liver epithelial cell line WB-F344 and their neoplastically transformed derivatives was evaluated as the major biomarker of chemopreventive effects. Set of nine selected phytochemicals or their analogues (diallyl sulfide, cinnamic acid, curcumin, emodin, indole-3-carbinole, metformin, quercetine, silibinin and thymoquinone) were tested at noncytotoxic doses for their ability to prevent inhibition of GJIC induced by environmental toxicants or tumor promoters in nontransformed WB-F344 cells: 1,1,1-trichlor-2,2-bis(4-chlorfenyl)ethan (DDT), fluoranthene, lindane, pentachlorophenol (PCP), perfluorooctanoic acid (PFOA) and 12 tetradecanoyl-13-forbolester (TPA). It was demonstrated that all these phytochemicals were able to prevent at least one of these chemicals from GJIC inhibition. The most pronounced effects were observed with quercetin and silibinin. Experiments with two selected chemicals, silibinin and metformin, showed, that these chemicals were able to increase GJIC in WB cells transformed with H-ras oncogene (WB-ras). Moreover, exposure to both metformin and silibinin induced apoptosis in WB-ras cell line, and metformin also increased the relative number of cells in the G2/M phase of cell cycle. These results provide novel information on chemopreventive and anticancer effects of studied phytochemicals and also indicate that in vitro evaluation of GJIC in WB cell lines represents a valuable tool for identification and characterization of chemopreventive and anticancer effects of chemicals.
