Macro-organization and flexibility of thylakoid membranes from Norway spruce

Abstract:

Cílem této bakalářské práce bylo vypracovat literární rešerši na téma makro-organizace thylakoidní membrány, její flexibilitu a principy spektrálně-optických metod, jejíchž použití nám poskytuje informace o makro-organizaci a flexibilitě thylakoidní membrány. V rešerši jsou shrnuty informace o složení lipidů a pigment-proteinových komplexů a jejich uspořádání v thylakoidní membráně. Teoretická část rovněž shrnuje základní informace o fotosystému I, fotosystému II a zabývá se procesy reorganizace, které přímo souvisí s fotoochrannými procesy vůči nadměrné ozářenosti. V experimentální části této práce byly využity dvě spektrálně-optické metody, cirkulární dichroismus a nízkoteplotní fluorescenční spektroskopie. V navržených experimentech byla měřena spektra cirkulárního dichroismu a nízkoteplotní fluorescenční spektra chlorofylu a na thylakoidních membránách smrku ztepilého (Picea abies [L] Karsten), huseníčku rolního (Arabidopsis thaliana L. cv. Columbia) a ječmene setého (Hordeum vulgare L. cv. Bojos) v různém prostředí. Hlavním cílem této práce bylo provést a zvládnout výše uvedené metodiky tak, aby na základě zjištěných změn makro-organizace thylakoidní membrány vlivem přítomnosti Mg2+ a vlivem přítomnosti osmotika (sorbitolu) v roztoku, bylo možné stanovit klíčové faktory pro udržení optimální vzdálenosti mezi thylakoidními membránami smrku. Na základě provedených měření a následném vyhodnocení dat bylo zjištěno, že u smrku ztepilého se závislost makro-organizace pigment-proteinových komplexů v thylakoidní membráně na koncentraci Mg2+ výrazně neliší v porovnání s ostatními studovanými rostlinnými druhy. Vliv osmotika na makro-organizaci thylakoidní membrány u téhož rostlinného druhu je zanedbatelný ve srovnání s kontrolními rostlinnými druhy. Ze získaných dat vyplývá, že pokud budeme snižovat koncentraci MgCl2 dojde k rozvolnění gran a LHCII následně přenese více energie na fotosystém I. Rovněž byl potvrzen význam kationtů Mg2+ pro granální strukturu a jeho stabilizaci. Sorbitol má vliv na makro-organizaci thylakoidní membrány, naopak minimální až zanedbatelný vliv na distribuci energie mezi fotosystémem II a fotosystémem I.

Abstract:

The aim of this bachelor's thesis was to prepare a literature search on the topic of macroorganization of the thylakoid membrane, its flexibility and the principal spectral-optical method, the use of which gives us information about macroorganizations and flexibility of thylakoid membranes. The research summarises information on the composition of lipids and pigment-protein complexes and their arrangement in the thylakoid membrane. The theoretical part further summarises the basic information about photosystem I, photosystem II and considers the processes of reorganization, which are directly related to photoprotective processes against excessive irradiation. In the experimental part of this thesis, two spectral-optical methods were used, circular dichroism and low-temperature fluorescence spectroscopy. In the proposed experiments, the spectra of circular dichroism and low-temperature fluorescence spectra of chlorophyll and on the thylakoid membranes of Norway spruce (Picea abies [L] Karsten), field beetle (Arabidopsis thaliana L. cv. Columbia) and barley (Hordeum vulgos L. c.) in different environments. The main goal of this work was to perform and master the above methodologies so that based on the changes in the macroorganization of the thylakoid membrane due to the presence of Mg2+ and the presence of osmotic (sorbitol) in solution, it is possible to determine key factors for maintaining optimal distance between thylakoid membranes of spruce. Based on the performed measurements and subsequent evaluation of the data, it was found that in Norway spruce the dependence of macro-organization of pigment-protein complexes in the thylakoid membrane on Mg2+ concentration does not differ significantly in comparison with other studied plant species. The effect of the osmotic agent on the macro-organization of the thylakoid membrane in the same plant species is negligible compared to control plant species. The data show that if we reduce the concentration of MgCl2, the gran will loosen and LHCII will subsequently transfer more energy to the photosystem I. The importance of Mg2+ cations for the granular structure and its stabilization was also confirmed. Sorbitol has an effect on the macro-organization of the thylakoid membrane, on the contrary, a minimal to effect on the energy distribution between photosystem II and photosystem I.
