Detection of honeybee pathogens using immunoanalytical methods

Abstract:

This bachelor thesis examines immunochemical detection of honeybee pathogens Melisococcus plutonius and Paenibacillus larvae, which are responsible for European and American foulbrood, respectively. Conjugates of specific antibodies with biotin and horseradish peroxidase were prepared and tested in enzyme-linked immunosorbent assay (ELISA). Afterwards, the assay parameters were improved by replacing enzyme labels with photon-upconversion nanoparticles (UCNPs) in the upconversion-linked immunosorbent assay (ULISA). The limit of detection (LOD) of the ULISA of 800 CFU ml-1 for P. larvae detection represented an improvement compared to the ELISA assay (LOD of 1000 CFU ml-1). In the case of M. plutonius, the LOD was 4×104 CFU/mL and 1000 CFU/mL in the case of ULISA and ELISA, respectively. The achieved results proved the potential of UCNPs as immunoassay labels. Finally, the analysis of real samples of bees, larvae, and bottom hive debris was successfully performed, demonstrating the practical potential of the ELISA and ULISA.

Abstract:

Tato bakalářská práce se zabývá imunochemickým stanovením včelích patogenů Melissococcus plutonius (původce hniloby včelího plodu) a Paenibacillus larvae (původce moru včelího plodu). Byly připraveny konjugáty protilátek s biotinem a křenovou peroxidázou, ověření jejich funkčnosti proběhlo pomocí enzymového imunostanovení (ELISA). Pro zlepšení parametrů imunostanovení bylo enzymové značení následně nahrazeno foton-upkonverzními nanočásticemi (UCNPs) v metodě upconversion-linked immunosorbent assay (ULISA). V případě metody ULISA bylo při stanovení P. larvae dosaženo limitu detekce 800 CFU/ml, což představuje zlepšení proti metodě ELISA (limit detekce 1000 CFU/ml). V případě bakterie M. plutonius byl limit detekce 4×104 CFU/ml případě ULISA stanovení a 1000 CFU/ml případě metody ELISA. Výsledky tak prokázaly vhodnost UCNPs jako značky v imunostanoveních. Nakonec byl praktický potenciál metod ELISA a ULISA demonstrován při analýze reálných vzorků včel, larev a měli.
