Application of chimeric antigen receptor (CAR) T-cells for solid tumors therapy

Abstract:

Chimerický antigenní receptor (CAR), je umělý receptor, který umožňuje T‐lymfocytům cílit na libovolnou antigenní strukturu na povrchu cílových buněk. Při vhodném výběru antigenu tak mají potenciál rozpoznat nádorovou buňku a účinně ji eliminovat. Po úspěších při zavedení této terapie vůči leukemiím a lymfomům, se výzkum také zaměřuje na terapie solidních nádorů. Tato diplomová práce se zabývá studiem CAR T‐lymfocyty vůči povrchovým antigenům solidních nádorů, neuroblastovému antigenu GD2 a prostatickému antigenu PSMA. Hlavním cílem práce je zavést účinný postup pro výrobu a expanzi CAR T‐lymfocytů. Vyrobené CAR T‐lymfocyty budou charakterizovány a jejich funkčnost bude testována in vitro na buněčných liniích. Výsledky experimentů pomohou optimalizovat výrobní postupy v nově založeném Centru buněčného a tkáňového inženýrství (CTEF‐CGMP) při Mezinárodním centru klinického výzkumu ve Fakultní nemocnici u sv. Anny v Brně (FNUSA‐ICRC).

Abstract:

Chimeric antigen receptor (CAR), is a manufactured receptor, which allows recognition of any antigenic structure on the surface of target cells. With the appropriate antigen selected, they can target and effectively eliminate tumour cells. After success with applying this therapy to leukaemias and lymphomas, the next target seems to be solid tumours. The focus of this work will be the neuroblastic antigen GD2 and prostatic antigen PSMA. The main goal is to establish an effective method for production and expansion of CAR T‐cells. The manufactured CAR T‐cells will be characterised, and their function tested in vitro using cell lines. Results of those experiments will help optimise production methods in the newly established Cell and Tissue Engineering Facility (CTEF) at the International Clinical Research Centre of St. Anne's University Hospital (FNUSA‐ICRC).
