Determination of bile acids in an automatic biochemical analyzer by two methods

Abstract:

Bakalářská práce se zabývá zpracováním literární rešerše na téma stanovení žlučových kyselin a jejich významu v klinické oblasti. Především je zaměřena na porovnání dvou komerčně dostupných laboratorních testů, které jsou založeny na principu fotometricko- enzymatického stanovení. Vyhodnocením získaných dat bude ověřena také stabilita reagencií při měření vzorků. Výsledky stanovené stávající používanou soupravou firmy Diazym na analyzátorech AU5800 firmy Beckman Coulter byly statisticky porovnávány s měřeními získanými využitím kitu firmy Dialab. Soupravy využívají reagencií, které jsou citlivé na světlo. Tyto kity byly verifikovány s využitím kontrolních (Seronorm human N = 12; Seronorm high N = 12; Biorad1 N = 12; Biorad2 N = 12; Biorad3 N = 12 a firemní interní kontroly Diacon N, N = 20 a Diacon P, N = 20) a pacientských vzorků (N = 147). Bylo ověřeno, že obě metody jsou citlivé na světlo a čím více se reagencie blíží konci své životnosti ve stroji, tím více se tato skutečnost projevuje v pravdivosti i preciznosti kontrolních materiálů. Na základě regresní analýzy a diferenciální grafické metody srovnávající měření žlučových kyselin 147 vzorků pacientů bylo zjištěno, že obě metody poskytují shodné výsledky.

Abstract:

The bachelor thesis deals with the processing of a literature review on the topic of the determination of bile acids and their importance in the clinical area. Above all, it focuses on comparing two commercially available laboratory tests based on the principle of photometric-enzymatic determination. The evaluation of the obtained data will also verify the stability of the reagents when measuring the samples. The results of the existing Diazym kit on Beckman Coulter AU5800 analyzers were statistically compared to measurements obtained using the Dialab kit. The kits use light sensitive reagents. These kits were verified using control (Seronorm human N = 12; Seronorm high N = 12; Biorad1 N = 12; Biorad2 N = 12; Biorad3 N = 12 and Diacon N, N = 20 and Diacon P, N = 20) and patient samples (N = 147). It has been proven that both methods are light sensitive and the more the reagents come to the end of their life time in the machine, the more this fact is manifested in the accuracy and precision of the control materials. Based on the regression analysis and differential graphical method comparing bile acid measurements of 147 patient samples, it was found that both methods provide consistent results.
