Isolation and identification of content compounds from selected plant material II

Abstract:

Diplomová práce se zabývá izolací a identifikací obsahových látek frakcí získaných předchozí separací extraktů rostliny Paulownia tomentosa (Thunb.) Steud. (Paulowniaceae), která obsahuje především flavonoidy, a to zejména prenylované povahy. Teoretická část práce shrnuje základní charakteristiku rostliny, popisuje obsahové látky a jejich biologickou aktivitu. V rámci experimentální části jsou popsány použité chromatografické metody k získání čistých látek, konkrétně tenkovrstvá chromatografie, sloupcová chromatografie a vysokoúčinná kapalinová chromatografie analytická i preparativní. Celkem bylo zpracováno 6 frakcí a získáno 18 látek s čistotou dostačující pro identifikaci pomocí metody společného nástřiku. Během této analýzy bylo úspěšně izolováno a identifikováno celkem 7 látek, jedná se o 3′-O-methyl-5′-hydroxydiplakon, tomenton B, paucatalinon C, paucatalinon I, diplakon, mimulol a pravděpodobně 4′,5,5′,7-tetrahydroxy-6-[6-hydroxy-3,7-dimethyl-2(E),7-octadienyl]-3′-methoxyflavanon. Zbývající frakce, u kterých se totožnost nepotvrdila, jsou vhodné pro zpracování jinými identifikačními metodami.

Abstract:

The diploma thesis focuses on the isolation and identification of the content compounds in fractions obtained through the previous separation of extracts from the plant Paulownia tomentosa (Thunb.) Steud. Paulowniaceae), which mainly contains flavonoids, especially prenylated flavonoids. The theoretical part of the thesis summarizes the basic characteristics of the plant, describes its chemical compounds and their biological activity. In the experimental section, the chromatographic methods used to obtain pure substances are described, specifically thin-layer chromatography, column chromatography, analytical and preparative high performance liquid chromatography. In total, 6 fractions were processed, and 18 compounds were obtained with sufficient purity for identification using the common spray method. During this analysis, a total of seven compounds were successfully isolated and identified, namely 3′-O-methyl-5′-hydroxydiplacone, tomentone B, paucatalinone C, paucatalinone I, diplacone, mimulol and 4′,5,5′,7-tetrahydroxy-6-[6-hydroxy-3,7-dimethyl-2(E),7-octadienyl]-3′-methoxyflavanone. The remaining fractions, for which identity was not confirmed, are suitable for processing using further identification methods.
