Detection possibilities of apoptosis in hyperbaric in vitro model

Abstract:

Apoptóza, tj. programovaná buněčná smrt (PCD) je u mnohobuněčných organismů fyziologický proces, uplatňující se po celou dobu života. Je důležitá z hlediska vývoje či obrany organismu, projevuje se morfologickými či biochemickými markery a zahrnuje složitou kaskádu reakcí. Hyperbarická oxygenoterapie (HBOT) je jedna z možností adjuvantní terapie bakteriálních infekcí měkkých tkání. Metoda je založená na inhalování 100% kyslíku za zvýšeného tlaku. Zvýšený parciální tlak kyslíku má řadu účinků, např. přímo působí na bakteriální kmeny, napomáhá hojení a regeneraci tkání. I přes značné klinické zkušenosti s touto metodou, zatím neexistují validní metodické postupy a modely HBOT. Z hlediska existence externích faktorů (antibiotika,HBO, aj.), které mohou mít výrazný vliv na viabilitu bakterií, buněk či tkáňových kultur, je důležité najít vhodnou metodu pro studium těchto účinků. Cílem této práce byla identifikace a zhodnocení vhodných metod k detekci apoptózy eukaryot a prokaryot, dále v kontextu HBOT. Na základě dostupných elektronických vědeckých článků byly metody k detekci apoptózy vybrány, definovány a posuzovány z hlediska použitelnosti, náročnosti a jiných parametrů. Můžeme konstatovat, že způsobů detekce apoptózy eukaryot je více než u prokaryot. Univerzální metodou se zdá být průtoková cytometrie a metoda TUNEL.

Abstract:

Apoptosis, ie. programmed cell death (PCD), is a physiological process in multicellular organisms that is applied throughout life. It is important for the development or defense of the organism, manifests itself in morphological or biochemical markers and includes a complex cascade of reactions. Hyperbaric oxygen therapy (HBOT) is one of the options for adjuvant therapy for bacterial soft tissue infections. The method is based on inhalation of 100% oxygen at elevated pressure. Increased oxygen partial pressure has a number of effects, eg directly affecting bacterial strains, helping healing and tissue regeneration. Despite considerable clinical experience with this method, there are no valid methodologies and HBOT models yet. In view of the existence of external factors (antibiotics, HBO, etc.) that may have a significant effect on the viability of bacteria and cells / tissue cultures, it is important to find a suitable method for studying these effects. The aim of this work was to identify and evaluate suitable methods for the detection of apoptosis of eukaryotes and prokaryotes, further in the context of HBOT. Based on available electronic scientific articles, methods for detecting apoptosis have been selected, defined and assessed for usability, difficulty and other parameters. We can conclude that the methods for detecting eukaryotic apoptosis are greater than for prokaryotes. Flow cytometry and the TUNEL method seem to be a universal method.
