Functional interactions of Mdm2 and MdmX proteins and molecular mechanisms affecting their levels in normal and cancer cells

Abstract:

Mdm2 and MdmX proteins, which are essential negative regulators of the p53 tumor suppressor, represent the crucial part of the normal healthy cells. Moreover, they are able to dramatically contribute to neoplastic cell transformation. This process can be caused by deregulation of their physiological activity or by an excessive increase of their cellular levels. Thorough understanding of their function and regulation can elucidate mechanisms in oncogenesis and bring progress in clinical trials. This thesis reviews the latest findings within this field focusing on the Mdm2 and MdmX structure and functions, and substances affected by their ubiquitin-ligase activity – primarily the p53. Furthermore, mechanisms of regulation, overexpression and other modifications impacting their gene expression are described in detail. Key processes include gene amplification, single nucleotide polymorphisms, alternative splicing of precursor mRNA as a source of the extended transcriptome, various post-translational modifications and many more interactions within the complex cell environment.

Abstract:

Proteiny Mdm2 a MdmX jako hlavní negativní regulátory tumor-supresorového proteinu p53 jsou důležitými molekulami nejen v zdravých buňkách, ale mohou mít významný vliv i na neoplastickou transformaci buněk. K té může přispět narušení fyziologické aktivity Mdm2 a MdmX, nebo nadměrné zvýšení jejich hladiny v buňce. K lepšímu pochopení procesů onkogeneze a případnému pokroku v klinické praxi je proto nezbytné důkladně porozumět funkci a regulaci těchto dvou proteinů. Předkládaná práce představuje souhrn nejnovějších poznatků na toto téma počínaje strukturou a funkcí proteinů Mdm2 a MdmX a molekulami ovlivněnými jejich ubikvitin-ligázovou aktivitou, především p53. Dále jsou detailně představeny mechanismy regulace daných proteinů, zvýšení jejich množství a různých změn již na úrovni genové exprese. Mezi klíčové děje patří především znásobení počtu kopií daných genů, jednobodové polymorfismy, různé posttranskripční úpravy jako alternativní sestřihy rozšiřující transkriptom, ale i množství posttranslačních modifikací a interakcí s jinými látkami v komplexním buněčném prostředí.
