Immunohistochemical localization of mitogen activated protein kinases in response of alfalfa (M. sativa) to elicitors

Abstract:

Teoretická časť tejto práce popisuje charakteristiku, význam a využite Medicago sativa L. Ďalej sa práca zaoberá mitogénom aktivovanými proteín kinázami (MAPK), ktoré slúžia ako signálne kaskády zodpovedné za prenos podnetov vyvolaných biotickým alebo abiotickým stresom. Podrobne je popísaný mechanizmus tejto signálnej kaskády a jej dôsledky na vývoj rastliny. Ďalej sa práca zaoberá stresovo-indukovanou MAPK (SIMK) M. sativa a zapojením MAPKs v imunite rastlín. Následne je uvedená charakteristika a mechanizmus pôsobenia elicitorov na rastliny. Elicitory sú látky, ktoré u rastlín vyvolávajú podobné obranné odpovede, ktoré sú spúšťané pri infekcii spôsobenej patogénom. Konkrétnejšie popisujeme elicitor flagelín, ktorý je súčasťou bičíku u gram-negatívnych baktérií. V experimentálnej časti sme pomocou nepriamej somatickej embryogenézy regenerovali rastliny divokého typu M. sativa kultivar RSY a tiež transgénnej línie exprimujúcej SIMKK RNAi. Sledovali sme vplyv biotického stresu, navodeného syntetickým elicitorom flg22, na lokalizáciu a abundanciu SIMK a fosforylovaných MAPKs v epidermálnych bunkách meristematickéj zóny koreňa pomocou imunohistochemickej metódy. Z výsledkov experimetálčasti vyplýva, že na intenzitu fluorescenčného signálnu SIMK mala najväčší vplyv 30 min inkubácia koreňov tekutým Fahräeus médiom.

Abstract:

The theoretical part of this bachelor thesis defines characteristic, importance a usage of Medicago sativa L. Further the thesis describes the role of mitogen-activated protein kinases which form important cascades to transfer stimulus created by biotic and abiotic stresses. We characterize the mechanism of this signalling cascade and what consequences MAPKs have on the development of plants. Additionally, the thesis defines the stress-induced MAPK (SIMK) and the involvement of MAPKs in plant's immunity. Subsequently it's specified the mechanism of function of elicitors on plants. Elicitors are substances which induce similar defensive responses in plants that are triggered by infections by pathogens. We describe in detail elicitor flagellin which is part of the flagellum of gram-negative bacteria. In experimental part we regenerated plants of wild type M. sativa of cultivar RSY and also transgenic line expressing SIMKK RNAi via indirect somatic embryogenesis. We studied the effect of biotic stress induced by the synthetic elicitor flg22 on the localization and the abundance of SIMKs and phosphorylated MAPKs in epidermal cells of the meristematic root zone using an imunohostochemical method. Our results show that the intensity of fluorescent signal SIMK was most affected by a 30 min incubation of the roots with the liquid Fahräeus medium.
