Production and characterisation of glyceraldehyd-3-phosphate dehydrogenase

Abstract:

Glyceraldehyde-3-phosphate dehydrogenase is a classical enzyme of glycolysis that promotes glucose metabolism. Its major task is to convert glyceraldehyde-3-phosphate to 1,3-bisphosphoglycerate. This process occurs in the cytoplasm. However, studies have shown that this enzyme is also found in the nucleus or in the cell wall. GAPDH on the surface of the cell wall is capable of adhesion. It can adhere to proteins of the extracellular matrix, especially to fibronectin and laminin. It is able to interact with plasminogen in the extracellular matrix and degrade it. This adhesive ability of GAPDH on the surface of bacteria or fungi is an important virulence factor. This thesis is focused on the summary of some pathogenic attributes of certain bacteria and fungi, especially on characteristics and adhesive ability of GAPDH from Candida albicans. The practical section is a sequel of a previous thesis where a construct containing the gapdh gene from Candida albicans, was prepared and inserted into organism Escherichia coli. The aim of this bachelor thesis is to test and optimize the conditions for protein production and purification.

Abstract:

Glyceraldehyd-3-fosfátdehydrogenasa je klasický enzym glykolýzy, který napomáhá metabolizmu glukosy. Jeho hlavní úlohou je přeměnit glyceraldehyd-3-fosfát na 1,3-bisfosfoglycerát. Tento proces probíhá v cytoplazmě. Výzkumy však dokázaly, že tento enzym se nachází v jádře i v buněčné stěně. GAPDH na povrchu buněčné stěny je schopný adheze. Dokáže adherovat na proteiny extracelulárního matrixu, hlavně na fibronektin a laminin. Taktéž je schopný interagovat s plasminogenem v extracelulárním matrixu a degradovat ho. Právě tato adhezivní schopnost GAPDH na povrchu baktérií nebo hub je významným faktorem virulence. Tato práce je zaměřená právě na shrnutí patogenních vlastností některých baktérií a hub, především na vlastnosti a adhezivní schopnost GAPDH z Candida albicans. Praktická část navazuje na předchozí práci, kde byl připravený konstrukt obsahující gen gapdh z Candida albicans, který byl vpravený do organizmu Escherichia coli. Cílem týto bakalářské práce je testovat a optimalizovat podmínky pro produkci proteinu a jeho purifikaci.
