Evaluation of antimutagenic effects of the plant extracts using the Ames test

Abstract:

Diplomová práce je zaměřena na charakteristiku vybraných rostlinných extraktů, které jsou stále častěji používány jako léčivé látky proti oxidačnímu, mutagennímu popř. karcinogennímu poškození buněk. V teoretické části práce jsou popsány sloučeniny získávané z rostlin, které mohou mít účinky prospěšné pro zdraví např. polyfenoly, antokyany, karotenoidy nebo vitamíny. Dále jsou podrobněji prostudovány farmakologické vlastnosti konkrétních rostlinných látek (salidrosid, allicin, vitamín C a kurkumin) a je popsán přístup k hodnocení jejich antimutagenních účinků. V experimentální části práce byla nejdříve ověřena metodika Amesova testu na standardní látce, kterou byl peroxid vodíku. Amesův test byl proveden s využitím indikátorového bakteriálního kmene Salmonella typhimurium TA102. Poté byly hodnoceny mutagenní a antimutagenní účinky extraktu z Aronia melanocarpa L., vitamínu C a kurkuminu.

Abstract:

Diploma thesis is focused on the characteristics of selected plant extracts which are more and more used as medicinal substances against oxidation, mutagenic, eventually carcinogenic cell damages. In the theoretic part, there are described compounds acquired from the plants, which might be beneficial to health, such as polyphenols, anthocyanosis, carotenoids and vitamins. Additionally, there were studied the pharmacologic features of specific plant substances in details (salidroside, allicine, vitamin C and curcumin) including the evaluation of their antimutagenic effects. In the practical part of the diploma thesis, the methodology of Ames test of hydrogen peroxide, being used as a standard substance, was initially verified. The Ames test was executed with the indicator bacterial strain of Salmonella typhimurium TA102. Afterwards, there were evaluated mutagenic and antimutagenic effects of Aronia melanocarpa L., vitamin C and curcumin extracts.
