The fnrP regulon of the bacterium Paracoccus denitrificans and its response to oxidative stress

Abstract:

Práce se zabývá vlivem transkripčního regulátoru FnrP na geny energetického metabolismu bakterie Paracoccus denitrificans. Studované geny byly vybrány na základě bioinformatické kontroly na přítomnost Fnr boxu. Výzkum proběhl především na úrovni transkripce pomocí RT-qPCR. Srovnáván byl divoký kmen a mutantní kmen s delecí FnrP, a to za normálních podmínek a prostředí oxidativního stresu, který by měl způsobovat rozpad FnrP a simulovat jeho deleci.

Abstract:

The thesis is focused on the impact of the FnrP transcription regulator on energy metabolism-related genes of the Paracoccus denitrificans bacterium. The studied genes were chosen based on bioinformatic confirmation of presence of the Fnr-box. The research was performed mostly at the level of transcription using RT-qPCR. The wild-type strain was compared to FnrP-deficient mutant strain in normal conditions and in conditions of oxidative stress, which should have caused a destruction of the FnrP and simulate its deletion.
