Interaction of cytokinins and cold stress

Abstract:

Působení nízkých teplot má velký vliv na produkci plodin a způsobuje nemalé ztráty zemědělcům. Tato práce byla zaměřena na vliv cytokininů a jejich signalizaci v odpovědi rostliny Arabidopsis thaliana na nízkou teplotu. V experimentální části byly využity linie s různou kombinací mutací v genech cytokininové signalizace AHP. Již za běžné teploty byla potvrzena dominantní role proteinů AHP2, AHP3 a AHP5, jejichž současná mutace vedla k výrazné retardaci růstu. Aplikace nízké teploty vedla k zastavení růstu plochy růžice u všech rostlinných linií. Na nízké teplotě však nebyl zastaven nárůst hmotnosti rostlin. Analýza transkriptu prokázala vliv proteinů AHP1, AHP2, AHP3 a AHP5 na regulaci genové exprese CBF po působení chladového stresu na rostliny. Dalšími analyzovanými geny byly regulátory odpovědi cytokininové odpovědi ARR-A, u kterých bylo po ošetření chladem pozorováno snížení (ARR4, ARR7) i zvýšení (ARR3) hladiny transkriptu, a to v závislosti na přítomnosti proteinů AHP. Vliv proteinů AHP byl nalezen i v regulaci exprese genů CRF, které byly dříve spojeny s působením nízké teploty. Nově bylo zjištěno, že na nízkou teplotu odpovídá i gen CRF1, ale pouze u linie s mutací v dominantních proteinech AHP. Tato práce potvrzuje a rozšiřuje znalosti o významu proteinů AHP v odpovědi rostlin na nízkou teplotu.

Abstract:

The effects of low temperatures have a high impact on crop production and cause significant losses to farmers. This work was focused on the role of cytokinins and their signalling in the cold response of Arabidopsis thaliana and its mutant lines. At standard temperatures, the dominant role of the AHP2, AHP3, and AHP5 proteins was confirmed by significant plant growth retardation. The cold treatment led to an inhibition of the growth of the rosette area in all plant lines. However, the increase in plant weight was not completely stopped at the low temperature. Transcript analysis showed the effect of the AHP1, AHP2, AHP3, and AHP5 proteins on the regulation of CBF gene expression after cold treatment. Next, the analysis of the expression of cytokinin response regulators revealed a decrease in the transcript level of ARR4 and ARR7, and an increase in the transcript level of ARR3 after cold treatment in an AHP-dependent manner. AHP proteins were also found as regulators of the gene expression of CRF genes that were previously described as responsive to low temperatures. Newly, we have identified the CRF1 gene to be responsive to low temperatures but only in the background with the mutations in the dominant AHP proteins. This work confirms and deepens knowledge about the importance of AHP proteins in the response of plants to low temperatures.
