Modification of platanic acid at position C-29

Abstract:

Tato bakalářská práce se věnuje modifikacím kyseliny platanové v poloze C-29 sousedící s ketonem. Nejprve bylo využito bromace této polohy, následně substituce bromu azidovým aniontem a výsledná 20-azidoplatanová kyselina byla dále využita pro sérii reakcí s různými alkyny za vzniku substituovaných triazolů (Huisgenova cykloadice). Vznikla tak série konjugátů kyseliny platanové s alifatickými, aromatickými i heterocyklickými substituenty, výhodná pro biologické testování a porovnání vlivu jednotlivých substituentů na celkovou aktivitu molekul. Teoretická část se zabývá obecně terpeny, triterpeny a kyselinou platanovou, a navíc zahrnuje přehled jejich derivátů, princip Huisgenovy cykloadice a sloučeniny triterpenů připravené touto reakcí v minulosti. Experimentální část se věnuje přípravě kyseliny platanové, optimalizaci její bromace, následné substituci azidem a v neposlední řadě optimalizaci podmínek Huisgenovy cykloadice. Bylo připraveno celkem pět nových konjugátů kyseliny platanové, jejichž struktura byla charakterizována pomocí NMR, HRMS a IČ. Také se testovala jejich cytotoxická aktivita na osmi nádorových a dvou kontrolních nenádorových buněčných liniích.

Abstract:

This Bachelor's thesis deals with a modification of platanic acid at the position C-29 next to the ketone group. Firstly, a bromination of platanic acid at this position was done, then a substitution of bromine with azide anion. The resulting 20-azidoplatanic acid was used for series of reactions with various alkynes which lead to a preparation of substituted triazoles (Huisgen cycloaddition). The new series of derivatives was prepared with aliphatic, aromatic and heterocyclic substituents, which was convenient for a biological assay and a comparison how the substituent affects the molecule's overall activity. A theoretical part covers an introduction of terpenes, triterpenes and platanic acid. It also provides an overview of platanic acid derivatives, a principle of Huisgen cycloaddition and an overview of triterpene derivatives prepared by this reaction before. An experimental part deals with a preparation of platanic acid, an optimization of its bromination, a following substitution with azide and an optimization of Huisgen cycloaddition. Five new compounds were prepared and characterized by NMR, HRMS and IR. They were also tested for cytotoxic activity on eight tumour cell lines and two control non-tumour cell lines.
