Host-specific parasites in interspecific hybrids of common carp and gibel carp

Abstract:

Hybridizace je v přírodě rozšířený fenomén. U máloostných se předpokládá výskyt hybridizace u 30% druhů, proto představují vhodný zdroj modelových druhů pro studium příčin a následků hybridizace. Z hlediska parazita představuje hybridizace novou proměnnou, která může zapříčinit genetické selhání hostitele a směřovat např. k vysoké abundanci parazita na hostiteli nebo naopak může u hostitele vést k heteróznímu efektu např. k snížení početnosti populace parazita nebo navodit rezistenci vůči parazitární infekci. Důsledky hybridizace byly testovány v liniích kapra obecného (Cyprinus carpio Linnaeus, 1758) a karase stříbřitého (Carassius gibelio Bloch, 1782), jejich reciprokých F1 hybridů, zpětných kříženců (4 linie) a F2 hybridech, získaných umělým výtěrem. Analyzována byla míra parazitární infekce zástupci taxonu Monogenea. Zaznamenáni byli zástupci rodů Dactylogyrus Diesing, 1850 (10 druhů) a Gyrodactylus von Nordmann, 1832 (7 druhů), počet parazitických druhů na linii ryb se pohyboval v rozpětí 6 až 12. Každá z linií rodičovských druhů vykazovala parazitaci hostitelsky-specifickými zástupci taxonu Monogenea. Abundance hostitelsky-specifických parazitů kapra obecného dosahovaly vyšších hodnot ve srovnání s abundancemi parazitů asociovanými s karasem stříbřítým. Linie F1 hybridů měly nižší míru parazitární infekce než rodičovské linie nebo vykazovaly rezistenci k hostitelsky-specifickým parazitům. Linie zpětných kříženců, vzniklé křížením F1 hybridů a kapra obecného, vykazovaly největší podobnost s kaprem obecným z pohledu abundancí hostitelsky-specifických parazitů tohoto druhu (zejména nejpočetnější Dactylogyrus extensus). Podobný trend byl zaznamenán i ve srovnání parazitace kapra obecného a linie F2 hybridů. Získané výsledky jsou v souladu s hybridní heterózou u F1 generací v případě infekce hostitelsky-specifickými zástupci taxonu Monogenea. V případě zpětných kříženců důležitou roli pro infekci hostitelsky-specickými parazity hraje proporcionální zastoupení jaderných genů v genomů hybridů. Možným vysvětlením vyšší míry parazitace u F2 hybridů je částečné genetické selhání.

Abstract:

Hybridization is a widespread phenomenon in nature. Hybridization is assumed to occur in 30% of the species of Cypriniformes, therefore this fish group represents a suitable source of model species for studying the causes and consequences of hybridization. From the point of view of the parasite, hybridization represents a new variable that can cause genetic failure of the host and lead, for example, to a high abundance of the parasite on the host or, conversely, can lead to a heterosis effect in the host, e.g. can cause a reduction of the parasite population or induce resistance to parasitic infection. The consequences of hybridization were tested using artificial produced lines of common carp (Cyprinus carpio Linnaeus, 1758) and gibel carp (Carassius gibelio Bloch, 1782), their reciprocal F1 hybrids, backcrosses (4 lines) and F2 hybrids. The level of parasitic infection by representatives of the taxon Monogenea was analyzed. Representatives of the genera Dactylogyrus Diesing, 1850 (10 species) and Gyrodactylus von Nordmann, 1932 (7 species) were recorded, the number of parasite species per fish line ranged from 6 to 12. Each of the parental species lines was parasited by host-specific monogenean species. Abundances of host-specific parasites on common carp reached higher values compared to abundance of parasites associated with gibel carp. F1 breed lines of hybrids had lower level of parasite infection than parental lines or showed resistance to host-specific parasites. Backcrosses lines, originated from crossing F1 hybrids and common carp, showed bigger similarity with common carp from the point of view of abundance of host-specific parasites of this species (especially the most abundant Dactylogyrus extensus). A similar trend was also found when comparing common carp and the line of F2 hybrids. The obtained results are consistent with hybrid heterosis in F1 generations form the point of view of infection intensity by host-specific monogenes. In the case of backcrosses, the proportional representation of nuclear genes in the genomes of the hybrids plays an important role for infection by host-specific parasites. A possible explanation for the high intensity of infection in F2 hybrids is partial genetic breakdown.
