Effect of functional modification of carbosilane dendrimers on complexation with siRNA

Abstract:

Tato diplomová práce se zabývá studiem potenciálního využití tzv. karbosilanových dendrimerů jako vektorů pro dopravu siRNA do buněk. Dendrimery se řadí mezi globulární nanopolymery. Tyto molekuly jsou charakteristické vysokou variabilitou vlastností. Ty jsou determinované především velikostí dendrimerů a povrchovými modifikacemi, které ovlivňují způsob interakce s molekulami biologických systémů. Specifické vlastnosti a schopnost dendrimerů vytvářet komplexy s biomakromulekulami na základě elektrostatických interakcí řadí dendrimery mezi nanočástice s potenciálem využití v mnoha sférách biomedicíny. Jednou z oblastí je jejich využití jako tzv. nevirových vektorů pro dopravu léčiv, terapeutických nukleových kyselin a oligonukleotidů do buněk. V této práci jsem věnovala pozornost přípravě a charakterizaci komplexů různých, zcela nových typů karbosilanových dendrimerů s siRNA. Vzniklé komplexy, tzv. dendriplexy jsem charakterizovala prostřednictvím několika metod, mezi něž patří elektroforéza, DLS, Zeta-potenciál, fluorescenční anizotropie a test ochrany siRNA před RNázou. Výsledky ukazují na vysoký potenciál využití těchto typů dendrimerů jako nevirových vektorů pro dopravu siRNA do cílových buněk a poukazují na významný vliv povrchových modifikací dendrimerů na výsledné vlastnosti dendriplexu.

Abstract:

This thesis deals with the study of the potential use, called carbosilane dendrimers as vectors to transport siRNA into cells. Dendrimers belong to the global nanopolymer. These molecules are characterized with high variability of feature. Their functional properties are primarily determined by the size of dendrimers and surface modifications that affect the way of the interaction with molecules of biological systems. The specific features and the ability of dendrimers to form complexes with biomacromolecules based on electrostatic interactions offer the potential applications in many areas of biomedical research. One of the areas is their use, as non-viral vectors for the transport of medicaments, therapeutic nucleic acids and oligonucleotides into cells. This thesis deals with the preparation and characterization of various complexes of completely new types of the carbosilane dendrimers with siRNA. Such complexes are called dendriplexes. I used to characterize them with several methods including electrophoresis, DLS, Zeta-potential, fluorescence anisotropy and the nuclease-resistence test. The results indicate a high potential to use of these types of dendrimers as non-viral vectors to transport siRNA into the target cells and show an important influence of surface modifications on the final feature of the dendriplex.
