Inaktivace nádorového supresoru p53 je významným bodem v nádorové transformaci mnoha typů buněk. Nejčastěji jí bývá dosaženo mutacemi v genu pro p53, ale u některých typů nádorů, které nesou nemutovaný p53, je za inaktivaci p53 zodpovědná zvýšená exprese onkogenu Mdm2, která vede k ubikvitinylaci p53 a k jeho nadměrné degradaci prostřednictvím proteazomu 26S. Abstract:
Inactivation of the p53 tumour suppressor is a significant point in the oncogenic transformation of many cell types. This transformation is the most frequently achieved by mutations in the p53 gene. However, in some types of tumour cells that retain wild-type p53, over-expression of the Mdm2 oncogene is responsible for inactivation of p53 and it leads to p53 ubiquitination and excessive degradation …more
