The role of the CD36 gene in the pathogenesis of Alzheimer's disease

Abstract:

Alzheimerova choroba je multifaktoriální neurodegenerativní onemocnění vyskytující se převážně u starších osob. Projevuje se poruchou kognitivních funkcí, ztrátou neuronů a viditelnými změnami na mozku. Na rozvoj choroby má největší vliv věk, dále úroveň vzdělání, kognitivní trénink a přidružená onemocnění, jako jsou kardiovaskulární choroby nebo úrazy hlavy a mozku. Receptor CD36 je membránový glykoprotein vyskytující se na celé řadě buněk. Svojí schopností vázat různé ligandy se účastní několika fyziologických ale také patologických procesů v lidském těle. V patogenezi Alzheimerovy choroby se uplatňuje vazbou Aβ, tvorbou zánětlivé reakce a aktivací oxidačního stresu. Cílem diplomové práce byla asociační studie polymorfismů rs1049673, rs1761667, rs1527483 genu pro CD36 s Alzheimerovou chorobou. Vybrané polymorfismy byly stanovovány u 983 osob české národnosti pomocí metody NGS. Při statistickém vyhodnocení získaných dat byla prokázána statisticky významná souvislost (p < 0,05) polymorfismu rs1049673 genu pro CD36 s bodovým hodnocením MMSE dotazníku. Statisticky významná souvislost (p < 0,05) byla prokázána také u polymorfismu rs1761667 genu pro CD36 s bodovým hodnocením MMSE dotazníku, věkem nástupu Alzheimerovy choroby a hodnotou BMI. Na základě získaných výsledků nebyl nalezen statisticky významný vztah mezi vybranými polymorfismy a Alzheimerovou chorobou (p > 0,05).

Abstract:

Alzheimer's disease is a multifactorial neurodegenerative disease occurring predominantly in the elderly population. It is manifested by impaired cognitive functions, loss of neurons and visible changes in the brain. The development of the disease is most influenced by age, then the level of education, cognitive training and related diseases, such as cardiovascular diseases or head and brain injuries. The CD36 receptor is a membrane glycoprotein found on a variety of cells. Due to its ability to bind various ligands, it participates in several physiological but also pathological processes in the human body. It is involved in the pathogenesis of Alzheimer's disease by the binding of Aβ, the formation of an inflammatory response and the activation of oxidative stress. The aim of the diploma thesis was the association study of polymorphisms rs1049673, rs1761667, rs1527483 of the CD36 gene with Alzheimer's disease. Selected polymorphisms were determined in 983 persons of Czech nationality using the NGS method. In the statistical evaluation of the obtained data, a statistically significant association (p < 0,05) of the rs1049673 polymorphism of the CD36 gene with the point evaluation of the MMSE questionnaire was demonstrated. A statistically significant association (p < 0,05) was also demonstrated for the rs1761667 polymorphism of the CD36 gene with the MMSE questionnaire score, the age of onset of Alzheimer's disease, and the BMI. Based on the obtained results, no statistically significant relationship was found between selected polymorphisms and Alzheimer's disease (p > 0,05).
