New methods and approaches in protein analysis

Abstract:

V současné době se pro analýzu bílkovin používá velké množství metod. V této práci jsou popsané některé z klasických analytických metod, například RTG krystalografie, NMR, CD spektroskopie, fluorescence a další. Tyto metody jsou v laboratořích dobře zavedené a používají se k nejrůznějším analýzám. Elektrochemické metody mají potenciál zařadit se mezi klasické analytické metody. Tyto metody jsou velmi citlivé ke strukturním změnám v proteinech i při malém množství vzorku. Výhodou elektrochemických metod je možnost miniaturizace pracovních elektrod pro praktické využití při tvorbě čipů pro paralelní analýzy. Elektrochemická analýza proteinů navázaných na povrchu elektrod je vhodná pro získávání dalších informací o proteinech, které nelze zjistit při analýze pomocí klasických metod. V této práci jsme porovnávali elektrochemické metody s některými klasickými metodami. Výsledky elektrochemických analýz amyloidových peptidů, α-synukleinu a proteinu p53 použitých v této práci vykazovaly vysokou shodu s použitými klasickými metodami.

Abstract:

Recently, large amount of methods is used for analysis of proteins. In this thesis, we describe some of the analytical methods such as RTG crystallography, NMR, CD spectroscopy, fluorescence etc. These methods are well established in the laboratories and are used for various analyses. Electrochemical methods have the potential to be included in the widely used standard methods. Electrochemical methods are very sensitive to structural changes of the proteins even in small amount of sample. The advantage of the electrochemical methods is the possibility of miniaturization of the working electrodes to chips formations for parallel analyses. Electrochemical analysis of proteins adsorbed to the electrode surface is suitable for procuring new information about proteins, which can not be obtained by means of standard methods. In this thesis we compared the electrochemical methods with the standard ones. The results of the electrochemical analyses of the amyloids peptides, α-synuclein and tumor suppressor protein p53 reported in this thesis exhibited a high degree of correlation with the standard methods.
