Vývoj a testování nových typů peptidů pro uplatnění v terapii neuroblastomu

Abstract:

Terapeutické přístupy založené na cílících či penetračních peptidech mohou být využity pro selektivní a efektivní transport léčiv. U neuroblastomu, jednoho z nejčastějších a nejletálnějších solidních nádorů dětského věku, je známo několik proteinů (hNET, TrkB a další), jež mohou být využívány pro cílenou terapii. V této práci byly pomocí homologního modelování připraveny strukturní modely hNET, které byly dále využity pro návrh cílících peptidů. Připravené peptidy byly charakterizovány a testovány in vitro jakožto cílící ligandy pro nanomedicínu buněk neuroblastomu se zvýšenou expresí hNET. V další části práce byl testován penetrační peptid Novicidin pro transport komplexu Schiffovy báze a zinku do buněk karcinomu prostaty. Novicidin byl dále studován pomocí funkčních analýz založených na sestřihu a mutacích aminokyselin. Vliv na sekundární strukturu peptidu byl studován jak pomocí výpočetních metod (molekulární dynamika), tak pomocí cirkulárního dichroismu nebo infračervené spektroskopie s Fourierovou transformací.

Abstract:

Peptide-based therapeutics such as homing peptides and cell-penetrating peptides can be used for selective and effective drug delivery, respectively. In neuroblastoma, one of the most common and lethal tumors in children, several protein targets have been implemented for targeted therapy such as hNET, TrkB and others. In this work, structural models of hNET protein were constructed via homology modeling and were used to design two homing peptides. The homing peptides were characterized and tested in vitro for applications of drug delivery via nanocarrier to hNET-expressing neuroblastoma cell lines. Further, a cell-penetrating peptide called Novicidin was used for delivery of a Schiff base-Zinc complex to prostate cancer cell lines. Functional analysis via peptide trimming and mutating was used to study Novicidin structure in more detail. The characterization of peptide secondary structure was done by several methods that complement each other, such as molecular dynamics (computationally), circular dichroism spectrometry and Fourier transform infrared spectroscopy.
