The evolution of asexuality in space and time on the case of European spined loaches of genus Cobitis

Abstract:

The ubiquity of theoretically inefficient sexual reproduction remains a major puzzle of evolution. Many ideas have been proposed to resolve this puzzle and provide sexual reproduction with some theoretical benefit over much rarer asexuality. Leading theories include the accumulation of deleterious mutations in clones (Muller's ratchet, Kondrashov's hatchet), or arms race with co-evolving parasites (the Red Queen hypothesis). However, empirical evidence to support these ideas is inconclusive. We have explored some of these ideas within the context of the Cobitis sexual-asexual complex, where sexual species coexist and compete with their asexual sperm-dependent hybrids. We have found that albeit asexuals are rather rare in nature, asexual hybrids can be a common phase during the speciation of sexual species, and historically, many more asexual taxa likely existed at some point in time. We have also tested the predictions of mutation accumulation in asexuals. Although mutations clearly accumulate in asexual genomes, with the highest mutation loads in the oldest clones, we have observed no trend towards more deleterious or more radical mutations, as predicted by the theory. We were able to rule out polyploidy and gene conversions as mechanisms compensating against mutation accumulation. Moreover, our tests of fitness-related phenotypic traits showed no degradation of older clones compared to young clones. Even though the role of gene conversions in alleviating mutation accumulation was negligible, they do seem to be adaptive, as we found several trends related to ploidy, subgenome ancestry, sequence composition, gene and allelic expression levels as well as function of affected genes.

Abstract:

Všudypřítomnost teoreticky nevýhodného pohlavního rozmnožování zůstává velkou hádankou evoluce. Bylo navrženo mnoho myšlenek k objasnění této hádanky a poskytnutí nějaké teoretické výhody sexuality nad mnohem vzácnější asexualitou. Vedoucí teorie zahrnují akumulaci škodlivých mutací v klonech (Mullerova rohatka, Kondrashovova sekyrka) nebo závody ve zbrojení s koevolvujícími parazity (hypotéza červené královny). Nicméně empirické důkazy na podporu těchto myšlenek nejsou definitivní. Prozkoumali jsme některé z těchto myšlenek v kontextu sexuálního-asexuálního komplexu Cobitis, kde sexuální druhy koexistují a soutěží se svými asexuálními sperm-dependentními hybridy. Zjistili jsme, že i když jsou asexuálové v přírodě vzácní, asexuální hybridi mohou být běžným stupněm při vzniku nových sexuálních druhů a historicky pravděpodobně existovalo mnohem více asexuálních taxonů. Také jsme otestovali predikce o hromadění škodlivých mutací v asexuálech. I když se mutace zřetelně hromadí, s největší zátěží pozorovanou u nejstarších klonů, nepostřehli jsme žádné trendy směrem k více škodlivým nebo radikálním mutacím, jak teorie predikují. Byli jsme schopni vyloučit polyploidii a genové konverze jako možné mechanismy kompenzující hromadění mutací. Navíc naše testy znaků biologické zdatnosti neodhalily žádné stopy degradace starších klonů oproti mladým klonům. Přestože genové konverze měly nevýznamný vliv na hromadění škodlivých mutací, zdají se být adaptivní jinak, jak ukazují nalezené trendy související s ploidií, původem subgenomů, kompozice sekvencí, genové a alelické exprese i funkce zasažených genů.
