Determination of ascorbic acid concentration by the method of cyclic voltammetry

Abstract:

Tato bakalářská práce se zabývá kvantitativním stanovením kyseliny askorbové. Všechny vzorky byly analyzovány metodou cyklické voltametrie pomocí přístroje EcaStat s využitím pracovní skleněné uhlíkové elektrody. Kyselina askorbová byla nejprve měřena ve třech různých vitamínových doplňcích. Měření bylo provedeno metodou kalibrační křivky a metodou přídavku standardu. Metodou kalibrační křivky byly zjištěny koncentrace 1105,5 mg/tab (Dr. Max Vitamin C 1000 mg citron 20 šumivých tablet), 258,6 mg/tab (Dr. Max Vitamin C 250 mg 100 tablet) a 105,9 mg/tab (Celaskon 100 mg 40 tablet). Následně byly hodnoty z obou metod porovnány s hodnotami deklarovanými výrobcem. Další měření kyseliny askorbové bylo v plodech růže šípkové (Rosa canina) a rakytníku řešetlákového (Hippophae rhamnoides). Pro extrakci aktivní složky byly použity dva různé extrakční roztoky. Plody obou rostlin vykazovaly vysoké obsahy vitamínu C. U šípku byl naměřen obsah vitamínu C 853,0 mg/100g (extrakční činidlo 2% kys. šťavelová) a 850,7 mg/100g (extrakční činidlo 2% kys. meta-fosforečná). U rakytníku byl naměřen obsah vitamínu C 504,9 mg/100g (extrakční činidlo 2% kys. šťavelová) a 585,5 mg/100g (extrakční činidlo 2% kys. meta-fosforečná). Výsledné koncentrace byly porovnány jak mezi sebou, tak i s údaji uváděnými v jiných studiích. Nakonec byl sepsán podrobný metodický návod na stanovení koncentrace kyseliny askorbové metodou cyklické voltametrie.

Abstract:

This bachelor thesis is concerned with the quantitative determination of ascorbic acid. All samples were analysed by cyclic voltammetry, using the EcaStat instrument, using a working glassy carbon electrode. Ascorbic acid was firstly measured in three different vitamin supplements. The measurements were carried out using the calibration curve method and the standard addition method. The calibration curve method yielded concentrations of 1105.5 mg/tab (Dr. Max Vitamin C 1000 mg lemon 20 effervescent tablets), 258.6 mg/tab (Dr. Max Vitamin C 250 mg 100 tablets) and 105.9 mg/tab (Celaskon 100 mg 40 tablets). The results from both methods were then compared with the values declared by the manufacturer. Further measurements of ascorbic acid were made in rosehip (Rosa canina) and sea buckthorn (Hippophae rhamnoides). Two different extraction solutions were used to extract the active component. The fruits of both plants showed high levels of vitamin C. For rosehip, 853.0 mg/100g (extraction reagent 2% oxalic acid) and 850.7 mg/100g (extraction reagent 2% meta-phosphoric acid) were measured. For sea buckthorn, 504.9 mg/100g (extraction reagent 2% oxalic acid) and 585.5 mg/100g (extraction reagent 2% meta-phosphoric acid) were measured. The resulting concentrations were compared with each other and with data reported in other studies. Finally, a detailed methodological guide for the determination of ascorbic acid concentration by cyclic voltammetry was written.
