The role of small leucin-rich proteoglycans in white/brown adipose tissue in humans

Abstract:

Tuková tkáň je tvořena buněčnými a nebuněčnými složkami, jenž se v průběhu celého života mění a přestavují. Tyto procesy jsou nezbytné při obezitě, která je charakterizovaná jako nadměrné rozšíření tukové tkáně. V tomto případě musí dojít k přestavbě extracelulární matrix (ECM), jelikož se zvětšují adipocyty a nadměrně se hromadí složky ECM, jako například kolageny či proteoglykany. Veškeré složky ECM se podílí na struktuře a funkci organismu a mohou mít i různé patogenetické úlohy. Tato práce se zabývá proteoglykany a jejich hlavními rolemi v organismu, převážně v ECM. Detailně se zaměřuje na skupinu malé proteoglykany bohaté na leucin (SLRPs). Konkrétně poskytuje seznam nejdůležitějších členů rodiny: dekorinu, biglykanu, asporinu, ECM2, lumikanu, osteoglycinu a podokanu, které se exprimují v tukové tkáni. Shrnuje zde důsledky, které vznikají při zvýšené či snížené expresi daných SLRPs. Zaměřuje se převážně na jejich role v tukové tkáni při obezitě, ale také na možné vztahy s onemocněními, které s obezitou souvisí.

Abstract:

The adipose tissue is generally composed of cellular and non-cellular components that change and rearrange throughout life. These processes are an inseparable part of obesity pathophysiology, whereas the whole process is characterized as an excessive expansion of adipose tissue. In this case, remodelling of extracellular matrix (ECM) always occurs because of enlarging adipocytes and excessive accumulation of ECM components, such as collagens or proteoglycans. All components of the ECM have both structural and functional aspects in the body and may have various pathogenetic roles. This thesis is focused on proteoglycans and their main roles in the body, especially in ECM. More specifically, the thesis is focused on the role of small leucine-rich proteoglycans (SLRPs). It gives the list of the most important members of the family: decorin, biglycan, asporin, ECM2, lumican, osteoglycin, and podocan, which are expressed in adipose tissue and elsewhere. Moreover, the pathophysiological consequences that arise from increased or decreased expression of given SLRPs in the body are discussed. Finally, the thesis focuses on the role of SLRPs in adipose tissue in obesity as well as possible relationships with obesity-related diseases.
