Synthesis of bisquaternary inhibitors of acetylcholinesterase used as a profylactic treatment of poisoning of organophosphorous´s inhibitors

Abstract:

Tato studie je zaměřena na inhibitory acetylcholinesterázy. Cílem bylo připravit výše uvedené látky a otestovat je v jejich inhibičních schopnostech standardní metodou in vitro. Práce je rozdělena na čtyři části. První se týká obecné charakteristiky vitálně důležitých enzymů. Pojednává také o látkách způsobující jejich zablokování, klinickými projevy intoxikace NPL a její terapií. Druhá část popisuje použité metody v syntetické části. Následující kapitola se zabývá postupem syntézy jednotlivých biskvartérních látek Biochemická část ukazuje vliv koncentrace připravených látek na aktivitu AChE.

Abstract:

The study is focused on the acetylcholinesterase{\crq}s inhibitors. The aime of this study was to prepare the symetric bisquaternary inhibitors and to test their inhibitor abilities by in vitro experiments. The thesis is divided into 4 parts. The first one includes common characteristic of a crucial enzyme in the human body. It deals with substances that involve their blocking too, the klinical signs of nerve agent poisonings and the treatment. The second one describes used methods in synthetic part. The following one is engaged in the methods of the synthesis of the bisquaternary inhibitors. The biochemistry part proves the influence of inhibitor{\crq}s concentration on the acetylcholinesterase{\crq}s activity.
