Structural characterization of Picornavirales infection

Abstract:

Echovirus 18 (E18) patří do rodiny Picornaviridae a způsobuje aseptickou meningitidu, encefalitidu nebo myelitidu. Virion E18 se skládá z jednořetězcového RNA genomu obklopeného proteinovým kapsidem s ikosahedrální symetrií. Kapsidový protein VP1 obsahuje hydrofobní kapsu s malou hydrofobní molekulou zvanou "pocket factor“, která stabilizuje virus a reguluje vypouštění genomu. K zahájení infekce se E18 částice navazují na receptory na buněčném povrchu. Virus vstupuje do buňky pomocí endocytózy. Po acidikaci endozomu se enterovirová částice změní v aktivovanou částici. Genom je poté vypuštěn z virového kapsidu přes endozomovou membránu do cytoplazmy buňky. Nové viry jsou vypuštěny z buňky jejím prasknutím. Tato disertační práce popisuje rekonstrukce E18 virů reprezentujících většinu z výše zmíněných kroků ze životního cyklu viru: nativní virus, komplex s neonatálním Fc receptorem, aktivovaná částice, otevřená částice a prázdná částice. E18 navázaná k neonatálnímu Fc receptoru obsahuje dvě konformace "pocket factor“, plně zanořenou a částečně vypuštěnou. Z aktivované částice E18 vystavené kyselému pH se odtrhne jeden či více pentamerů z kapsidových proteinů. Díra v kapsidě po chybějících pentamerech je dostatečně velká pro vypuštění genomu. Obě tato nová pozorování určila enterovirové mezistavy, které vyplňují mezery v chápaní životního cyklu enterovirů a mohou pomoci při vývoji antivirotik.

Abstract:

Echovirus 18 (E18) is from the family Picornaviridae and causes aseptic meningitis, encephalitis, or myelitis. The virion of E18 is composed of a single-stranded RNA genome, surrounded by a protein capsid with icosahedral symmetry. The capsid protein VP1 forms a hydrophobic pocket, which contains a small hydrophobic molecule called a pocket factor that stabilizes the virus particle and regulates genome release. To initiate infection, E18 particles attach to receptors at the cell surface and enter the cell by endocytosis. After acidication of the endosome, enterovirus virions transform into activated particles. Activated particles release their genomes, which are translocated across the endosome membrane by an unknown mechanism. New virions are released by the bursting of an infected cell. This thesis describes reconstructions of E18 particles, representing most of above-mentioned virus life cycle steps: native virion, complex with neonatal Fc receptor, activated particle, open particle, and empty particles. E18 bound to the neonatal Fc receptor contains two conformations of pocket factor: fully buried and partially expelled from the pocket. Activated particles of E18 exposed to acidic pH expel one or more pentamers of capsid protein. The hole in the capsid from the missing pentamer is sufciently large for genome release. Both of these observations describe novel enterovirus genome-release intermediates, which fill gaps in the enterovirus life cycle and may enable the development of new antivirals.
