Determination of lipid-peroxidation products during defence reaction

Abstract:

V průběhu obranné reakce u rostlin dochází k aktivaci procesu peroxidace lipidů. Tento děj je vyvolán účinkem enzymů lipooxygenas či reaktivních forem kyslíku. Vede k degradaci polynenasycených mastných kyselin membránových lipidů. Vznikají tak strukturně rozmanité látky, které v rostlině plní obranné, signální či stimulační funkce. V důsledku své vysoké reaktivity však mohou i poškozovat buněčné molekuly. Primárními produkty peroxidace lipidů jsou hydroperoxidy mastných kyselin. Jejich hladina a jednotlivé zastoupení vypovídají o celkové peroxidaci lipidů v rostlině. Představují prekurzory sekundárních produktů, které vznikají jejich radikálovým rozpadem či účinkem enzymů v oktadekanové dráze. Cílem praktické části práce bylo porovnat heterologní expresí produkovaný kryptogein (X24) a jeho mutantní formu L41F ve schopnosti vyvolat peroxidaci lipidů v listech rostliny Nicotiana tabacum. Míra peroxidace lipidů byla posouzena na základě množství vzniklých hydroperoxidů mastných kyselin. Hydroperoxidy byly analyzovány po redukci na příslušné hydroxy mastné kyseliny metodou HPLC. Získané výsledky poukázaly na velmi nízkou schopnost mutantní formy kryptogeinu L41F vyvolat peroxidaci lipidů. X24 naopak vykazoval silnou nekrotickou aktivitu s indukcí především 9-hydroperoxidů mastných kyselin. Provedená optimalizace metody pro stanovení hydroxy mastných kyselin metodou HPLC vyústila v použití dvou nových podmínek, které byly porovnány s již zavedenou metodou.

Abstract:

During plant defense response process lipid peroxidation is activated as a result of the effect of enzymes lipoxygenases or reactive oxygen species. Lipid peroxidation leads to the degradation of polyunsaturated fatty acids presented in membrane lipids. Moreover, it leads to production of structurally diverse compounds that play roles in defense, signal or stimulative functions. However, due to their high reactivity these compounds could cause damage to cell biomolecules. Primary products of lipid peroxidation are fatty acid hydroperoxides. Levels and proportional representations of these compounds in plants indicate lipid peroxidation profile. They act as precursors of secondary products that are formed from primary compounds by radical decomposition or by the effect of enzymes involved in octadecanoic pathway. The aim of the practical part of this work was to compare the ability of heterologous expression prepared cryptogein (X24) and its mutant form L41F to induce lipid peroxidation in leaves of Nicotiana tabacum. The lipid peroxidation level was evaluated according to the amount of produced fatty acids hydroperoxides, which were analyzed after their reduction to corresponding hydroxy fatty acids by HPLC. Obtained results pointed out very low ability of mutant form of cryptogein L41F to induce lipid peroxidation. On the contrary, X24 exhibited strong necrotic activity mainly with fatty acids 9-hydroperoxide induction. Performed method optimization of hydroxy fatty acids determination by HPLC resulted in practical use of two new conditions, which were compared with original established method.
